Objective:To observe the effects of dexmedetomidine pretreatment to prevent Lidocaine induced convulsion in New Zealand white rabbits and effects on neurotransmitters. Methods:forty male New Zealand white rabbits,2-2.5kg,were divided randomlyinto 4 groups,with 10 rats in each group;C group:physiological saline;group D1:Pre injection of physiological saline+Convulsion induced by lidocaine;group D2:3 ?g / kg pre injection of dexmedetomidine + lidocaine induced convulsion;group D3:5 ?g / kg pre injection of dexmedetomidine +lidocaine induced convulsion.Group C:Pump into 0.9% physiological saline(15 min);group D1-3 were pumped into the different concentrations of dexmedetomidine with liquid 8ml(10 min),Continuous pump with 2% lidocaine at the rate of 4mg/kg/min,stop pump at the moment of Tonic convulsion occurred.recorded convulsion appeared time(T),Duration of convulsion(TT).Measure the content of plasma lidocaine?excitatory amino acids in cerebrospinal fluid(aspartate(Asp),glutamate(Glu)) and inhibitory amino acid(glycine(Gly), gamma amino acid(GABA)) at the time of base value(T0), Tonic convulsion occurred(T1), 30 min After convulsion(T2). Results:D1-3 groups in New Zealand rabbits were convulsions,compared to D1 group,D2? D3 group appear convulsion time was significantly prolonged(P<0.05), lidocaine concentrations increased(P<0.05);compared with the D2,D3 group appeared seizure was prolonged(P<0.05);D1,D2,D3 group convulsion duration had no significant difference(P<0.05).Compared with T0, D1-3 group T1,T2 time excitatory amino acids were increased,decreased glycine(P<0.05).Compared with the D1 group,the excitatory amino acids in D3 and D2 groups decreased,glycine increased,and the excitatory amino acids in D3 group increased less than D2 group(P<0.05). Conclusion:A certain dose of dexmedetomidine preset can prevent the effect of convulsion induced by lidocaine with dose effect relationship.The preventive effect may be related to the inhibition of excitatory amino acids increased.
