Objectives To observe the effect of Huoxue Tongluo Lishui Recipe on the expression of excitatory amino acid and inhibitory amino acid neurotransmitters in rabbit vitreous fluid in the model of acute retinal ischemia-reperfusion induced by anterior chamber perfusion,and to explore the therapeutic effect and possible mechanism of Huoxue Tongluo Lishui Recipe on retinal edema,so as to provide theoretical basis for clinical treatment of ischemic retinal diseases.Methods Twenty-two New Zealand rabbits weighing 2-2.5kg,half male and half female,were randomly divided into two groups,11 in the model group and 11 in the administration group.The rats in the administration group were given Huoxue Tongluo Lishui recipe(21.3g/kg/d)twice a day,and the rats in the model group were given saline of equal volume as the control group.1.Six RIRIRI models were established in the model group and the administration group respectively.Optical coherence tomography(OCT)was performed at each time point before and 6,12,1,2,3 and 5 days after the establishment of the model to accurately measure retinal thickness.2.Five microdialysis probes were implanted into the vitreous body from the model group and the administration group,and the isotonic phosphate buffer was used for 1.5 ?l/min.After 60 minutes of perfusion rate balance,the RIRIRI model was collected once every 20 minutes and three times as selfcontrol.After 120 minutes,the RIRI model was made.The dialysate was collected continuously from the beginning of the model establishment and once every 20 minutes.Finally,the excitability and inhibition of rabbit retina were detected by high performance liquid chromatography(HPLC)from the dialysate collected before the model establishment,during the ischemia process and at different time points after reperfusion.Trends in amino acid levels.The data were analyzed by SPSS 20.0 statistical software.Results 1 OCT examination showed that the retinal interlayer structure was clear and the boundary was obvious before the model was made,and the retinal structure was gradually disordered and blurred after the model was made.The retinal thickness increased gradually from 6h to 3D after modeling,and reached the maximum at 1D.At 5th day after modeling,the retinal thickness decreased to the level before modeling(P > 0.05).Statistical analysis showed that the retinal thickening degree of the drug group was weaker than that of the model group 6 hours to 3 days after modeling,and the difference was statisticallysignificant(P < 0.05).2.The results of HPLC showed that Asp in the model group increased temporarily at the beginning of high intraocular pressure,and then returned to normal.Glu in vitreous fluid of the model group increased sharply 10 minutes after ischemia,then decreased to baseline level,and increased significantly again 10 minutes after reperfusion,reaching 289.2% of baseline level,and then decreased to baseline level again.Glu level increased again after 3 hours of reperfusion,reaching 643.7% of baseline level at 290 minutes of reperfusion.Glu level also increased slightly in the administration group,but the increase was lower than that in the model control group,reaching 330.2% of the baseline level at 290 min of reperfusion.At 270,290 and 310 minutes of reperfusion,the Glu level in the administration group was significantly lower than that in the model control group(P < 0.05).The levels of Gly,Tau and GABA in vitreous fluid of model control group and traditional Chinese medicine administration group did not change significantly,and there was no significant difference between the two groups at each time point.Conclusions Huoxue Tongluo Lishui Prescription has obvious therapeutic effect on retinal edema caused by retinal ischemia-reperfusion injury.Its mechanism may be to inhibit the toxicity of excitatory amino acids by reducing the excessive release of Glu in the retina,thus exerting the protective effect of retinal ischemia-reperfusion.At the same time,it has no significant effect on the levels of Asp,Gly,Tau and CABA.Figure27;Table13;Reference 91...
