Objective:Because of ischemia and hypoxia,the body releases a large number of inflammatory cytokines after traumatic shock.However,giving fluid resuscitation treatment is easy to be injured by ischemia and reperfusion.At present,the protective effect of low hypothermia on organ has been gradually obtained.This study through the method(wigger 's modified method)of femur fracture and traumatic bloodletting to establish the traumatic hemorrhagic shock model,to study the effects of cold exposure on inflammatory cytokines in rats after resuscitation from traumatic shock.Medthod:1.Establishment of traumatic shock model and grouping of animals:Severty-two adult SD male rats,weighting 300-350 g,were randomly divided into ambient-sham operation group,hypothermia-sham operation group,hypothermia-traumatic shock group and ambient-traumatic shock group.Four groups were further divided into three subgroups including 8 hour group,24 hour group and 36 hour group.The animals were anesthetized by intraperitonea injection of chloral hydrate after weighted,using the method of femur fracture and traumatic shock bloodletting to establish the traumatic shock model.The sham operation group only underwent bilateral carotid artery and vein catheterization.Rats in each group after recovery from resuscitated were placed in the appropriate temperature observing 8h,24 h,36h.2.Results record: Recording rectal temperature of each group and killing the rats at the end of the observation time.The levels of cytokines of IL-6,TNF-?,IL-1? and IL-10 in the serum,heart and lung were measured by Enzyme linked Immuno Sorbent AssayRusults:1.Changes of rectal temperature in rats:When the hypothermia trauma group was placed in the low temperature environment,temperature temporarily decreased,then gradually rose at about 6h.It maintained at the mild hypothermia 28?-35? during0-8h.The constant temperature level was maintained at 37?-38 ? after 16 h.2.The concentration of inflammatory cytokines IL-6,TNF-a,IL-1,IL-10 in serum and heart lung tissue : At 8h point,comparing hypothermia-traumatic group with the ambient-traumatic group,the expression of inflammatory cytokines of IL-6,TNF-?and IL-1? in serum,heart and lung tissue was significantly lower(p<0.05).The expression level of inflammatory cytokines IL-10 was significantly increased(P<0.05).However,the opposite result was appeared at 36 h,the differences were statistically significant(p<0.05).Conclusion:1.After resuscitation to the traumatic hemorrhagic shock,ischemia / reperfusion induced a strong inflammatory response in the body.2.At the early time exposured to low temperatures can significantly mitigatie inflammation reaction in the serum and tissue.3.The later will exacerbate serum and tissue inflammation,which might cause damage to heart and lung tissue.
