Objective To investigate the protective effect of hydrogen-rich HC-A kidney preservation solution on cold ischemia and reperfusion injury of kidney in rats.Methods Twenty-four healthy male Wistar rats were randomly divided into3groups. In Control group (Hi group), only right kidney was removed. Clod ischemia/reperfusion was performed in common kidney preservation solution group(H2group). By catheterizations in right renal artery and vein, perfusion and outflow tracts were set up. After cutting off abdominal aorta and postcava in the right position at the same time, the left kidney was perfused by4℃common HC-A kidney preservation solution. After cold perfusion, left kidney was stored in a container which was full of4℃common HC-A kidney preservation solution for60min. In hydrogen-rich kidney preservation solution group(H3group), the perfusion and preservation solution was the only difference compared with H2group, hydrogen-rich solution instead. After24hours, the venous blood was obtained to detect the levels of BUN, Cr, TNF-a and IL-6. Left kidney was removed to detect the levels of MDA and8-OHdG in tissue homogenate. The activity of caspase-3in renal tissue was also detected. The pathological changes of kidney and apoptosis of renal tubular epithelial cells were evaluated by light microscopy. The injury of renal tubular epithelial cells was estimated in Paller scoring method.Results The levels of MDA,8-OHdQ BUN, Cr, TNF-a, IL-6and the activity of caspase-3in H2and H3groups rised statistically compared with H1group (P<0.05) Compared with H2group, the levels of MDA,8-OHdG, BUN, Cr, TNF-a, IL-6and the activity of caspase-3reduced statistically in H3group (P<0.05). There were no obvious morphologic changes in Hi group. The injury of renal tubules in H2group was obvious, while it was slighter in H3group. In H2group, structures of kidney tubules were severely damaged, many epithelial cells were necrosis and fell off. While in H3group, the phenomenons above ameliorated. The Paller score in H2and H3groups rised statistically compared with H1group (P<0.05),while the Paller score in H3group was statistically lower than that in H2group (P<0.05). The apoptosis of renal tubular epithelial cells in H2group was obvious, while it was slighter in H3. The number of apoptotic cells per high power field in H2and H3groups rised statistically compared with H1group (P<0.05), while the number in H3group reduced statistically compared with H2group (P<0.05)Conclusion1. After cold perfusion and preservation, ischemia and reperfusion injury looks obviously. Oxidative stress, inflammatory response and apoptosis probably play an important part.2. Hydrogen-rich HC-A kidney preservation solution can ameliorate renal function of rats. Lipid peroxidation and oxidative damage of DNA were alleviated, while inflammatory cytokines’ release decreased. The injury and apoptosis of kidney tubule epithelial cells were also alleviated.
