Objective To study the effect of the dendritic cells(DCs)subsets infection on CCL-11 and IL-13R?2 expression and EOS infiltration in OVA-induced lung tissue cells by adoptive transfer of DC subsets from mice infected with Schistosoma japonicum.Methods DC subsets from mice infected with S.japonicun were respectively isolated and purified using CD8? and CDllc microbeads.Twenty BALB/c mice were randomly divided into four groups.Group normal served as the control.Group OVA was the asthma group.Each mouse in group SJCD8?~+ DC/OVA was injected with CD8?~+DC from mice infected S.japonicun,while each mouse group SJCD8?~-DC/OVA was injected with CD8?~-DC.One hour after adoptive transfer,All but group normal were sensitized and challenged with ovalbumin to induce asthma.Mice were sacrificed after four weeks and their lungs were removed for HE staining and immunohistochemistry staining,inflammatory changes were observed,and levels of CCL-11 and IL-13R?2 expression were detected in lung tissue.Results HE staining indicated that group normal had not inflammation and SJCD8?~-DC/OVA had slight inflammation in lungs.The groups SJCD8?~+DC/OVA and OVA had serious inflammation and larger number of infiltrating inflammatory cells in lungs.The mice of Normal?OVA?SJCD8a~+DC/OVA and SJCD8?~-DC/OVA lung tissue immuohistochemical staining showed,the average opitical density(AOD)of CCL-11 are 0.1523±0.0109,1.4967±0.2256,1.3435±0.0949 and 0.8634±0.2114,the positive area of CCL-11 in blood vessels and trachea's percentage of the total area are 0.0004±0.0003,0.2572±0.0859,0.2103±0.0231 and 0.0392±0.0054;the AOD of IL-13R?2 are 0.1527±0.0153,0.3345±0.0037,0.2832±0.0350 and 0.1755±0.0201,the percentage of the IL-13R?2's area of the total area are 0.0370±0.0341,0.2970±0.0062,0.2870±0.0310 and 0.1171±0.0050.The difference in percentage and AOD between groups was statistically significant(P<0.001).Conclusion The CD8?~-CD11c~+DC subset from mice infected with S.japonicun can inhibit the OVA-induced expression of CCL-11 and IL-13R?2 and inhibit asthma.
