Along with lifestyle changes, the prevalence of type 2 diabetic nephropathy is increasing dramatically in China, which caused great pressure to the social medical service. So the development of prevention and treatment methods for type 2 diabetic nephropathy should be accelerated. Medical nutrition therapy has become an indispensable therapy for diabetic nephropathy(nephropathy DN, diabetic) by adjusting the structure of nutrients. However, for patients, the intake of nutrition content, especially the type and content of protein, is difficult to be unified. This delays the development of special medical purpose foods. New form homogenated nutritional products, is a perfect vehicle for enteral nutrition preparation. This study uses application of biotechnology, food technology and modern instrumental analysis technology to develop a homogenate diet as the main form of special medicinal formula based on the basis of the animal experiment. The main results are as follows:1. A dominant diabetic nephropathy model was established. For model group, the level of 24 h urine volume, glycemic index, 24 hours microalbuminuria(24 h umalb), 24 hours urine protein(24 h UTP), urine albumin and urinary creatinine ratio umAlb/Cr were significantly higher than the level of nonpathogenic group, and their difference had statistical significance(P < 0.05).2. The evaluation of low protein binding plant protein diet to delay the development of diabetic nephropathy had been completed. Low protein group's angiotensin prime II, serum creatinine, fasting blood glucose, serum cystatin C, serum urea, serum triglyceride had significant difference with the standard's(P < 0.05). Two protein proteins of glutenin and peanut protein were tested, the two groups with casein group were decreased and had statistically significant in contrast to the above indexes(P < 0.05). Through kidney hematoxylin and eosin(HE) staining we found that in standard casein DN model group, glomerular volume increases, widened mesangial, matrix increased, basement membrane thickening, nearly convoluted tubular epithelial cells visible brush border disappeare, tubular vacuolization of epithelial cells, renal mass visible inflammatory cell infiltration, renal tubular epithelial cells slight degeneration and necrosis, and low protein group and valley histone kidney situation improved significantly.3. A special medical nutrition formula-homogeniazed diet which suitable for patients with diabetic nephropathy was developed. Improve the industrialization of homogenate meal of preparation method, overcome the shortcomings of the existing technical manual preparation and operation, by using high pressure homogenization technique and spraying drying technology of multi nutrient components of an embedding, the preparation process of the production greatly simplified, the production efficiency improved. Through the single factor experiment and the orthogonal experimental design, the high pressure homogenization homogenization pressure was 77 MPa, spray drying process of inlet air temperature was 185°C, wind temperature was 140°C. At 50°C for brewing temperature, 275 mL for water quantity, the diabetic nephropathy(DN) special medicinal formula in nutrition and physicochemical and hygiene have reached the quality standard requirements of patients was developed.
