Objective:To observe the effect of NO and ET-1 on the micro vascular endothelial cells(HUMCE)in the human heart of Fu Fang Teng to the hypoxia/oxygen(H/R)injury.Methods:The HCMEC was cultivated by artificial subculture in vitro,and the 3 rd generat-ion of HCMEC was inoculated into the DMEM-culture flask which was filled with pure nitrogen for 10 minutes in advance.Then,the flask was sealed after the air inside was eliminated by filling pure nitrogen again.After 2-hour cultivation in 5%CO2 incubator with 37?,the original DMEM was changed by the new one,and the flask was cultivated under the same condition again.Finally,the H/R model of HCMECs was established.Experiments were randomly divided into:Normal control group and model group(Fu Fang Teng group,Kato Purygroup,model control group).Uses the drug serum method to preparation of HUMCE containing serum on rat passaged culture intervention.Expression of ET-1 protein was detected by WB assay,NO concentration was determined by nitrate reductase method.Results:Compared with model control group,the Fu Fang Teng group,Kato Pury group and the protein levels of ET-1 in normal group were significantly decreased(P<0.05),NO secretion was significantly increased(P<0.05);Comparison with the Fu Fang Teng group,the captopril group ET-1 protein levels were significantly increased(P<0.05),NO secretion was significantly reduced(P<0.05).Conclusion:Fu Fang Teng not only can be raised the HUMCENO level after hypoxia/anoxia injury,but also cut ET-1 protein expression,maintain ET/NO balance,thus protecting the endothelial function of HUMCE.
