| Objectives: To explore the possible disorders of Th17/Treg cells factors as well as the relationship between serum 25-(OH)D3 levels and cytokine imbalance of Th17/Treg in patients with Hashimoto’s thyroiditis by means of measuring serum 25-(OH)D3,related cytokines of Th17 and Treg cells in Hashimoto’s thyroiditis patients and healthy controls.Methods: Thirty-eight patients with newly onset hypothyroidism or subclinical hypothyroidism on account of Hashimoto’s thyroiditis were included in this study as HT group,they had never been treated,all of them consecutively came from the Endocrinology Clinic of the Second Hospital of Hebei Medical University from May to August in 2015.Simultaneously,42 cases of normal control were recruited as NC group(Normal Control group).The two groups matched for age and sex.Serum free T3(FT3),free T4(FT4),TSH,TPOAb and TGAb were measured by means of Immunochemiluminometric assays.Serum 25-(OH)D3,IL-6,IL-10,IL-17 A,IL-23,TGF-β1 were measured by means of ELISA.All statistical data were dealt with SPSS19.0,and P<0.05 was considered statistically significant.Results:1 Between the two groups,serum 25-(OH)D3 levels,IL-10 levels,TGF-β1 levels in HT group were lower than that in NC group [(11.91±4.27)ng/ml vs.(15.66±4.81)ng/ml,(1.93/1.96)pg/ml vs.(2.94±1.56)pg/ml,(2988.47±605.64)pg/ml vs.(4615.17 ±1206.39)pg/ml],differences were statistically significant(P<0.05).Serum IL-17 A levels in HT group were significantly higher than that in NC group(there were only 6 samples in HT group measurable because of the sensitivity of human IL-17 A ELISA kit,they were 1.06pg/ml,1.58pg/ml,1.29 pg/ml,2.00pg/ml,0.61pg/ml and 1.91pg/ml,the concentrations of the remaining 32 patients in HT group and all of the 42 cases in NC group were below 0.55 pg/ml).Although serum IL-6 levels and IL-23 levels in HT group were higher than that in NC group [(1.54/3.08)pg/ml vs.(1.15/3.03)pg/ml,(1.82/9.40)pg/ml vs.(1.82/0.41)pg/ml],differences between the two groups were not significant(P>0.05).2 For objects in HT group,serum 25-(OH)D3 levels were negative correlated with TSH(r=-0.427,P=0.008),TPOAb(r=-0.689,P<0.001),TGAb(r=-0.428,P=0.007).Serum 25-(OH)D3 levels were positive correlated with FT3(r=0.396,P=0.014),FT4(r=0.349,P=0.032).Serum 25-(OH)D3 had no significant correlation with IL-6,IL-10,IL-23 and TGF-β1(P>0.05).Conclusion:1 Serum 25-(OH)D3 levels in Hashimoto’s thyroiditis group were significantly lower than that in normal controls.Cytokine of Th17/Treg disorders occured in patients with Hashimoto’s thyroiditis,with the levels of IL-17 A increased and the levels of IL-10,TGF-β1 decreased.2 Whether 25-(OH)D3 was involved in the pathogenesis of HT by affecting the Th17/Treg imbalance of cytokines remained to be further identified. |
