Objective: Hashimoto's thyroiditis?HT?is a common thyroid diseases in Endocrine field in recent years and the incidence rate is increasing year by year.It affects the quality of life of patients.The pathogenesis of HT,however,is still not very clear.According to the existing data,Helper T cells and 25?OH?D3 play an important role in the pathogenesis of Hashimoto's thyroiditis.T helper cells affect thyroid by secreting cytokines to regulate,and 25-?OH?D3 affect thyroid by cellular and humoral immunity.But it is currently unclear that whether there is a correlation between them.What is the secretion of various types of helper T cytokines and what is the relationship between thyroid function and thyroid related antibodies.In addition,what is the change of Th2 cytokines in the balance of Th1/Th2.About the above problems,there are only some of controversial and less comprehensive study based on the existing research we have now.So,we conducted this experiment that Hashimoto's thyroidit is the carrier and 25-?OH?D3 and thyroid function are the cores to discuss the relationship among them.Methods:1 This experiment divide the subjects into two groups,one is untreated Hashimoto's thyroiditis group?case group?,the other one is normal control group?control group?.All of the subjects were collected morning fasting venous blood to detect s-TSH,FT3,FT4,TPOAb,TGAb,IL-2,IL-4,IL-6,IL-10,IFN-?and 25-?OH?D3.2 All data are used SPSS 17.0 software for statistical analysis.It takes bilateral inspection to test,taking P<0.05 as statistical significance.Result:1 Comparison between the age,BMI,sex ratio,there were no significant differences between the case group and the control group?P>0.05?.2 The serum levels of IFN-?and IL-2 in the case group were significantly higher than those in the control group?P<0.05?and 25-?OH?D3 and IL-10 were significantly lower than those in the control group?P<0.05?.There was no significant difference between IL-6 and IL-4.3 The serum levels of 25-?OH?D3 in the case group were significantly lower than those in the control group?P<0.05?.25-?OH?D3 was negatively correlated with IFN-?and IL-2 and positively correlated with IL-4.25-?OH?D3 was no correlated with the levels of serum IL-6?IL-10.4 S-TSH,TPOAb and TGAb in case group were all negatively correlated with 25-?OH?D3 and positively correlated with IFN-?.Both s-TSH and TPOAb in case group were positively correlated with IL-2.5 The multiple stepwise regression analysis that s-TSH is dependent variable and serum IL-2 and IFN-?are independent variable showed that serum IFN-? significantly affect on the levels of s-TSH in case groups.Then s-TSH was changed to TPOAb,the result is 25-?OH?D3,too.Conclusion:1 Compared with the healthy controls,there is a remarkable lower serum IL-10 level and higher serum IL-2,IFN-?levels of the patients of HT.The outcome prompted that the upregulation of Th1 cytokines and downregulation of Th2 cytokines may be exist at the same time and Th1 cytokines affects the patients with HT more significantly than Th2 cytokines.But among Th1 cytokines,IFN-?affects the patients with HT the most significantly.2 The serum levels of 25-?OH?D3 in the case group were significantly lower than those in the control group?P<0.05?.25-?OH?D3 was negatively correlated with IFN-and IL-2 and positively correlated with IL-4.25-?OH?D3 was no correlated with the levels of serum IL-6?IL-10.The outcome prompted that the lower serum 25-?OH?D3 affect s-TSH through the upregulation of Th1 cytokines and downregulation of Th2 cytokines.
