BACKGROUND: With the rapid development of social economy and the improvement of people's living standard, more and more attention has been paid to the health problems. With the deepening the study of obesity people, more and more risk factors is gradually recognized. The study has confirmed that the pathogenesis of obesity is a complex process, is the result by genetic factors and environmental factors of long-term interaction, of which genetic factors is obesity students based on the intrinsic and environmental factors is the exterior condition of the obesity, which indicates that genetic factors may play an important role in the pathogenesis of obesity,and obesity susceptibility genes has become a hot topic in the current obesity research.OBJECTIVE: To explore the relationship between NNMT gene polymorphism and obesity, and to provide a new theoretical basis and target for the prevention and treatment of obesity.METHODS: The body composition test was conducted on male college students. According to the results of the body composition test and the questionnaire survey, 783 subjects were selected. The subjects were divided into high fat group and low fat group. Gene from the human genome project database(Project Database HapMap) to obtain the reference sequence of the NNMT gene and single nucleotide polymorphism information. According to the analysis of linkage disequilibrium, select nnmt gene of 19 single nucleotide polymorphism loci in association studies,polymerase chain reaction ligase detection reaction(PCR LDR) detection of SNP genotypes. At last, 151 subjects were screened for resting energy metabolism of different genotypes of different genotype.RESULTS: According to the student body size analysis, the percentage of body fat in high fat group was higher than that in low fat group, the percentage of body muscle after body weight calibration and resting energy metabolism was lower than that of low fat group, and the two had significant difference(P<0.05). Resting sites rs10891644 gene rs10891644 according to genotype analysis found that site gene type and allele gene frequencies in the high fat group and low fat group had significant difference(P = 0.002, allelic gene frequency P = 0.029). Further analysis found that inCO dominant genetic model genotype frequency distribution in the high fat group and low fat group two groups have significant difference(P < 0.01), the GT genotype of body fat percentage higher than GG and TT genotype, muscle percentage was than GG and TT genotype is low, and the difference is significant(P < 0.01); to exclude the effects of body weight of GG genotype energy metabolism is higher than that of GT and TT genotype, no significant difference(P > 0.05).CONCLUSION:(1) NNMT gene polymorphism is significantly associated with obesity, and the rsl0891644 variation has a significant effect on body composition.(2) from physiological analysis results, the lower the relative resting energy consumption rate(RREE=REE/ weight) is an important cause of obesity one,but variation of rsl0891644 and did not cause REE significant changes, visible rsl0891644 variation and not through regulation of REE in this way variant components.(3) from the perspective of behavioral analysis, rsl0891644 variation has a significant impact on diet and exercise habits, which may be the main factor in the change of body composition caused by rsl0891644 variation.
