Objective: To perform the correlation analysis between the chromatographic fingerprints and anti-inflammatory data of Abutilon indicum n-butanol extract,the study on serum pharmacochemistry and metabolic components in the urine,to clarify the therapeutic basis of Abutilon indicum n-butanol extract.Methods:(1)Experimental study of anti-inflammatory effect : Mouse ear swelling induced by xylene and cotton ball granuloma were adopted in this study to investigate the influence of Abutilon indicum n-butanol extract for acute or chronic inflammation.(2)Experimental study on fingerprint spectrum :HPLC method was used to compare and study on ten areas Abutilon indicum n-butanol extract.(3)Spectrum correlation analysis: Used Pearson analysis method of SPSS bivariate correlation,to perform the correlation analysis between the anti-inflam-matory data and 19 common chromatographic peaks in fingerprints of Abutilon indicum n-butanol extract come from 10 areas.(4)Study on serum pharmacochemistry : Half male and female SD rats to the high dose of 51.42 g raw/kg,once a day,oral administration of rats for 4 days,aortaventralis blood make in an hour after oral administration on the last day.According to the above method for HPLC chromatographic analysis,through the drug-containing serum compared with the original,blank serum chromateogram,judge moving points in the serum.(5)Analysis of metabolitecomponents in urine :Comparative analysis of urine samples after oral administration with the blank urine,drug-containing serum chromatogram with using HPLC method,judge the absorbed in the blood serum and the metabolic components of Abutilon indicum n-butanol extract.Results:(1)The ear edema induced by xylene(P<0.01),and weight cotton granuloma in mice(P<0.05)were inhibited significantly by Abutilon indicum extract come from ten areas.(2)Establishment a same pattern fingerprint of Abutilon indicum n-butanol extract,and calibrated a total of 19 peaks,10 areas of samples and the control fingerprint similarity are above 0.8.(3)There was a correlation between the 3 chromatographic peaks and the suppression of the swelling of the ear in mice.The 6 chromatographic peaks were some extent correlated with the inhibition of the mouse granuloma(P>0.05).The 8th chromatographic peaks have significant correlation with these two pharmacological effects(P<0.05).(4)The comparison and analysis between medicinal serum,raw materials and blank serum shows that there have 11 substances absorbed into blood,six of which is the prototype medicinal ingredients and another five are metabolites or is the product of rats under stress.(5)Urine samples show 7 components are detected,3 of which are the final metabolite absorbed into the blood,the other 4 may be multiple metabolic transformation of the product or the other substances.Conclusion:(1)The Abutilon indicum n-butanol extract have the inhibitive effects on the acute and chronic inflammation;(2)Fingerprints of 10 batches of Abutilon indicum n-butanol extract have good similarity with contrast fingerprint,but between each of them the similar is not high.There have chemical composition and content differences between eath places.(3)Multiple components together to produce anti-inflammatory effects in Abutilon indicum.(4)The serum transitional composition and metabolite componentsin urine mainly from three aspects: the original ingredients as well as its metabolite components of Abutilon indicum n-butanol extract or from the rats with under stress.
