Objective:To establish a model of the anti-oxidant and anti-inflammatory spectrum effect of Plantago asiatica,combining the research of"spectrum"and"effect",to clarify the material basis of Plantago asiatica’s anti-inflammatory and antioxidant effects,and to provide a scientific basis for its material basis and mechanism of action,Also laid the foundation for improving the quality evaluation system of Plantago asiatica.Methods:By DPPH method and ABTS method,using the IC50,EC50and vitamin C equivalent of the two free radical scavenging rates as indicators,the in vitro antioxidant activity of Plantago asiatica from different origins was studied;the RAW 264.7 cell inflammation model induced by LPS,Using the contents of inflammatory factors NO,IL-6 and TNF-αas indicators,the in vitro anti-inflammatory activity of Plantago asiatica from different origins was studied;the chromatographic conditions of the fingerprints of Plantago asiatica from different origins were investigated by high performance liquid chromatography to establish different origins Fingerprint of Plantago asiatica,and determine its common peak;through similarity evaluation and cluster analysis,screen the best Plantago asiatica batches for subsequent study on the relationship between spectrum efficiency;based on principal component analysis,gray correlation analysis and partial least squares Multiplying regression analysis method to establish the relationship of anti-oxidation and anti-inflammatory spectrum effect of Plantago asiatica.Results:The results of in vitro antioxidant activity showed that Plantago asiaticas from 10 different origins all had good scavenging ability on DPPH free radicals and ABTS free radicals,and the scavenging ability of free radicals was concentration-dependent.Among them,Psyllium from Guangdong Grass has the best effect on DPPH free radical scavenging,and Hebei Plantago has the best scavenging effect on ABTS free radicals;in vitro anti-inflammatory activity studies have found that Plantago from 10 different origins can inhibit the inflammatory factors NO,IL-6 and TNF.-αrelease,and the inhibitory effect is concentration-dependent.Plantago in Bijie,Henan and Sichuan has better anti-inflammatory activity than other origins;similarity evaluation and cluster analysis found that the results of the two are consistent,and the results are similar.Seven different producing areas of Plantago asiatica with a degree greater than 0.8 and cluster analysis belonging to the same category were used to carry out the follow-up spectrum efficiency relationship.The seven batches of producing areas were Shaanxi,Guangxi,Guizhou Bijie,Guangdong,Sichuan,Henan and Hebei;7 different producing areas were established.The fingerprint of Plantago asiatica has 19 common peaks identified,peak 6 is vanillic acid,peak 8 is psyllium glycoside,peak 9 is verbascum glycoside,peak 10 is Plantago asiatica D and 17 The peak is luteolin and peak 18 is apigenin,and the content of the above-mentioned designated peaks in Plantago asiatica from different origins is quite different;principal component analysis divides the 19 common peaks into five main components,the second,fifth,Peaks 6,11,13,15,and 17 are classified as principal components of the first category,peaks 3,9,10,12,and 16 are classified as principal components of the second category,and peaks 7,18,and 19 are classified as principal components of the second category.There are three types of principal components,peaks 8 and 14are classified as the fourth principal components,and peaks 1 and 4 are classified as the fifth principal components.The results of gray correlation analysis and partial least squares regression analysis show that,The main pharmacodynamic peaks of Plantago asiatica anti-oxidant activity are peaks 6,8,9,14,and 15.The main pharmacodynamic peaks of Plantago asiatica anti-inflammatory activity are confirmed to be peaks 4,6,11,and15.Conclusion:Based on the fingerprint analysis of Plantago asiatica from different origins,Plantago asiatica uses multiple components and multiple pathways to synergistically exert its antioxidant activity and anti-inflammatory activity.This study effectively analyzed the two active medicinal components,interpreting the medicinal material basis of Plantago asiatica’s antioxidant and anti-inflammatory effects to a certain extent,which is helpful for the quality control and the study of the mechanism of Plantago asiatica.
