Objective:This article investigated the effect of Tau on PTEN protein expression,and investigate Tau whether through P13K/AKT signal pathway to inhibit the proliferation of hypoxia-induced pulmonary artery smooth muscle cells (PASMCs).Methods:The original generation develop PASMCs in rats,uesd 2-5 generations in experiments.The concentration of Tau is 80 mmol/L. The concentration of LY294002 is 10 mmol/L.The concentration of Rapamycin is 10 mmol/L. ① Using MTT to determate the relationship of dose conversion and function.Western blot and immunofluorescence assay determate the effect of Tau on PTEN protein expression. The PASMCs were divided into 4 groups:normoxia group,normoxia+Tau group,hypoxia group,hypoxia+Tau group. ② Using westernblot and immunofluorescence and flow cytome-try(FCM) assay the effect of P13K/AKT signal pathway. The PASMCs were divided into 5 groups:normoxia group.hypoxia group,hypoxia+Tau group, hypoxia+Rapamycin group, hypoxia+Tau+Rapamycin group and normoxia group,hypoxia group,hypoxia+Tau group, hypoxia+LY294002 group, hypoxia+Tau +LY294002 group.Results:①MTT assay revealed that taurine(80 mmol/L) significantly inhibited PASMCs proliferation induced by hypoxia(p<0.05),but had no effect on PASMCs under normoxia condition. ②Tau could increase the expression of PTEN by hypoxia. ③ The expression of PCNA(p<0.01) and CyclinD (p<0.05) in hypoxia group were higher than that in normoxia group,which were significantly suppressed by taurine. There was no effect of Tau on the expression of P-AKT、AKT、mTOR.④ Tau had no effects on the expresstion of P-AKT(p<0.01) and mTOR(p<0.01) under normoxia condition,however,Tau decreased the expresstion of P-AKT and mTOR induced by hypoxia. Compared with hypoxia group,both LY294002 (10 μmol/L) and Tau (80 mmol/L) could decrease the expression of PCNA, CyclinD、P-AKT. In Tau+LY294002 group,the expression of PCNA,CyclinD and P-AKT were decreased much more than simply used Tau or LY294002. ⑤Flow cytometry results confirmed that, compared with the control group, Tau and LY294002 can prevent the hypoxia status of PASMCs into the S phase, so as to inhibit its proliferation.Conclusion:Tau could increase the expression of PTEN by hypoxia.Tau also could inhibit PASMCs proliferation induced by hypoxia and this maybe via P13K/AKT pathway.
