Objective:To observe the variation of VEGFm RNA and BMP2 m RNA as well as patho-change in the steroid induced avascular necrosis of femoral head under the effect of panax notoginseng saponins,Discussing the control efficiency and mechanism of panax notoginseng saponins on SANFH,To provide experimental basis for the prevention and treatment of SANFH with traditional Chinese medicine of notoginseng.Methods: 135 male New Zealand rabbits were used to establish the model of steroid induced avascular necrosis of the femoral head by using the improved horse serum combined with methylprednisolone.After the model was completed,the model was identified by using the correlation detection indicators such as pathological section.After the success of the model identification using random number table method were randomly divided into three groups,namely normal saline group,compound Ossotide group and Panax notoginseng saponins group.Were given normal saline,compound bone peptide and notoginseng total saponin in femoral head local targeted drug therapy.Dose of 0.1ml/kg/w,once a week,continuous treatment for four weeks.After the treatment,the air embolism method was put to death,Take the side of the femoral head through the femoral head HE staining of tissue morphology and empty bone lacuna rate detection.On the other side and ultra low temperature freezing,and the expression of VEGFm RNA and BMP-2m RNA real-time fluorescence quantitative PCR detection of femoral head.Experimental results using SPSS.22 statistical software for data analysis.Results:1.After the model rabbits were slightly depressed mental state,Dim,fluffy hair,activity is reduced,and the lower limb becomes awkward.loss of appetite,the size of normal,slightly reduced weight.HE dyeing observation visible trabecular bone thinning,femoral head fracture,bone cells scattered distribution is irregular,the tibial bone marrow cells decreased,fat cell hyperplasia,empty bone lacuna increased,confirmed that the model was successful.2.After treatment,the gross morphology and the observation of the femoral head were observed:(1)Physiological saline group:The spirit is dispirited,bleak color,unkempt hair removal,decreased activity,loss of appetite,weight loss was significantly,femoral head did not see obvious subsidence,lose luster is faded,grey on the whole,thinning of the cartilage surface of femoral head,femoral neck with osteoporosis,chisel cut easily.(2)Compound Ossotide group:Well mental state,color,fluffy but luster,no obvious removal,occasionally bounce,appetite is normal,weight has no obvious change.Rabbit femoral head form is complete,the surface luster is a bit dark,pale white on the whole,the femoral neck bone is hard,difficult to chisel cut.(3)notoginseng total saponin group:Good mental state,color luster a fluffy,bounce activities and normal appetite,weight increased slightly.Luster on the surface of the femoral head form is complete,and with pale white,femoral neck bone is hard,difficult to chisel cut.3.Groups of femoral head HE dyeing observation:(1)Physiological saline group:Bone tissue continuous middle,trabecular appear rupture,trabecular bone fibrosis and sequestrum formation,inter trabecular bone marrow hematopoietic cells decreased,bone cells scattered irregular,fat cells increased significantly,the empty osteocyte nest increased significantly,empty bone lacuna rate was 20.0%;(2)Compound Ossotide group:Bone tissue continuous fair,trabecular bone slightly thinner,trabecular fibrosis,visible new bone formation and bone cell distribution rules and intramedullary femoral head visible hematopoietic cells and fat cells without significantly increased,empty bone lacuna rate was 15.93%;(3)notoginseng total saponin group:Bone tissue continuous fair,trabecular bone slightly thinner,failure is not obvious,visible new bone formation and bone cell distribution rules,intramedullary femoral head visible hematopoietic cells and empty osteocyte nest no significant increased,empty bone lacuna rate was 15.10%.The ratio of empty lacuna results by variance analysis,Compound bone peptide and notoginseng total saponin group clear difference compared with normal saline group,respectively,are statistically significant(P<0.05).Compound bone peptide with notoginseng total saponin group was no significant difference,no statistical significance(P>0.05).4.The expression of VEGFm RNA and BMP-2m RNA in each group was detected by real-time fluorescence quantitative PCR.Compound bone peptide and notoginseng total saponin group,respectively,compared with normal saline group,the expression of BMP2 m RNA are statistically significant(p<0.05),Notoginseng total saponin group compared with compound bone peptide BMP2 m RNA expression of no statistical significance(p>0.05);Compound bone peptide and notoginseng total saponin group,respectively,compared with normal saline group,the expression of VEGFm RNA are statistically significant(p<0.05),And notoginseng total saponin group is better than that of bone peptide group(p < 0.05).Conclusions:1.Hormone can result in the expression of rabbit femoral head tissues VEGFm RNA and BMP2 m RNA significantly decreased,which affect the tissue repair of femoral head.2.Panax Notoginseng Saponins can enhance the expression of VEGFm RNA and BMP2 m RNA in the bone tissue of rabbits with steroid induced avascular necrosis of the femoral head.3.Panax Notoginseng Saponins has some prevention and cure function on the rabbit with steroid induced avascular necrosis of the femoral head.
