The Study Of Ghrelin To Improve The Microcirculation Disturbanceand Lipid Metabolism Disorder Of The Steroid-induced Avasular Necrosis Of Femoral Head In Rabbits

Objective:To observe the effect of Ghrelin to Vascular Endothelial Growth Factor, Bone Morphogenetic Protein-2, Triglycerides, Total Cholesterol, Low Density Lipoprotein and High Density Lipoprotein.To stuty microcirculation disturbance and disorder of lipid metabolism in rabbits with avascular necrosis of the femoral head,providing dates and some theoretical base for the therapy of avascular necrosis of the femoral head.Methods:Divide30male New-Zealand rabbits, which weight2.5-3.Okg, age24weeks into2groups randomly:26into model group and4into normal group. Horse serum combined with hormone to build model of steroid-induced avasular necrosis of femoral head.Except the deaths after successful models, dividing the left20rabbits into2groups:10into control group, another into intervention group.The intervention group injects Ghrelin (25ug/kg/day)2weeks, and the control group gives the same saline2weeks too.After2weeks, collecting blood, killing animals, collecting specimens, observing the histomorphology of femoral head, detecting the fasting lipid, testing the VEGF and BMP-2level of femoral head with Real-timePCR.Results:Horse serum combined with hormone can successfully build model of steroid-induced avasular necrosis of femoral head:from the MRI we can see flake long signals on T1weighted imaging,some long signals at T2weighted imaging and high signal on T2WI. In articular cavity we can fluid signal. On pathological section we can see empty lacuna increasing, bone marrow cells instead of fat, bone nucleus pyknosis in model group.The intervention group with Ghrelin in histomorphology shows:compared with control group, the empty lacuna increase, the adipocyte is more, bone trabecula become therarefaction., the adipocyte hypertrophic slightly, and the proportion is bigger than normal in marrow but no pisparity phenomenon.In the Ghrelin intervention group the level of TG, CHO and LDL is higher than the control group, but the HDL is lower, and the differences is statically significant (P<0.05).Also the expression of VEGF and BMP-2is higher than the control group, the differences is statically significan (P<0.05)Conclusion:Ghrelin can improve microcirculation of the femoral head, promote the bone formation by improving the lipid metabolism of avasular necrosis of femoral head and the expression of VEGF and BMP-2in femoral head tissue.
