Background: With the development of modern industry and transportation industry,spinal cord injury is not rare in modern society,and it will bring about serious symptoms and complications.Its occurrence rate is high,and it is often accompanied by autonomic dysfunction,its mortality rate is about 4.4%~16.7%.After spinal cord injury often to patients bring serious sequelae,in the damage to the spinal cord segments corresponding to the limb nerve dominate the region appears a series of motor and sensory dysfunction,often combined with sphincter dysfunction,caused by the size of obstacles,and muscle strength and muscle tone out abnormal abnormal and pathological reflex.Common sequelae of paraplegia and paralysis of the limbs,and it is more difficult to recover even along the patient's all the life.Although there are some patients with spinal cord injury after spinal cord injury,the motor and sensory function can be recovered,but the degree of recovery is very limited.At present,there is no effective treatment for patients with spinal cord injury,and the effect of various treatment measures can not reach the desired goal.The reason is difficult to restore nerve function after SCI,one of the important reasons is the damage zone of micro environment through a series of complex changes,generated a lot of inhibitory factors that inhibit axonal growth.Chondroitin sulfate protein polysaccharide is a very important axonal regeneration inhibitory factor,when after spinal cord injury CSPGs in synaptic transmission and spinal cord regeneration recovery process exhibit strong inhibition,inhibit the regeneration of axons and nerve function.While chondroitinase ABC can specifically degrade CSPG and don't damage other tissue.Previous research has confirmed BDNF can promote neuronal survival and differentiation through receptor kinases,so as to realize the cholinergic neurons in vivo or in vitro survival and differentiation,and ultimately play a role in promoting the repair of spinal cord injury.Objective: This topic through the modified Allen's method established rat model of spinal cord injury and subarachnoid catheter,and by ductal injection of ChABC and BDNF and study of BDNF(brain-derived neurotrophic factor)combined with ChABC(chondroitinase ABC)on spinal cord injury healing and repair of neural function of rats.Methods: The SD rats randomly divided into false operation group,saline treatment group,BDNF treatment group,ChABC treatment group and the BDNF combined ChABC treatment group,to establish a rat model of spinal cord injury.Through the BBB score,somatosensory evoked potential(SEP)and motor evoked potential(MEP)monitoring,respectively at 1 day after spinal cord injury in rats,7 days,14 days,28 days comparative observation of rat spinal nerve function recovery.Results: 1,All animal BBB score before operation was 21,sham operation group,postoperative scores did not change significantly,the rest of the group after injury of lower limbs in rats with complete paralysis,second days after injury gradually restored,hindlimb function had different degrees of recovery,3 ~ 5 group was significantly better than that of the group,on the 28 day hindlimb motor function recovery rate was the highest BBB score up to 6.4 points;2,Evoked potential showed that the combined treatment group on the 14 day of the incubation period of SEP is significantly shortened(p<0.05),on the 28 day incubation period of MEP compared with other groups significantly shortened(p<0.05),showed that the application of BDNF and ChABC combined treatment of rats with spinal cord injury can promote the recovery of neurological function,can reduce the damage of SEP and MEP in spinal cord of rats at early stage of incubation,SEP was obviously improved,late of MEP was improved significantly.3,Hematoxylin eosin(he)staining: all treatment groups had different degree gliosis and glial scar formation.Among them,the third,fourth,fifth group and the second group compared less proliferation of glial cells,gymnasium and II group,significantly reduced.Showed that application of ChABC and BDNF can injury to the spinal cord glial cells inhibits the increase in students,the United Application of BDNF and ChABC effect the best.4,Nissl staining : On the first day after the injury,the control group and the treatment group of Nissl bodies by normal tigroid structure gradually disintegrated,broken down into fine dust like particles,both dissolution of Nissl bodies.At about 1 week peaked.Starting from the second week,the Nissl body gradually restored in the form,reduce the fine dust like particles,scattered in patches,at the same time,coloring number of neurons gradually increased.Combination group than in the other treatment groups and control group.The results show that application of BDNF and ChABC combination therapy for promoting axonal regeneration and promote neuronal repair effect is better than single drug treatment group and control group.Conclusion: There was neuroprotective and neurotrophic effect by BDNF combined with ChABC on spinal cord injury,the proliferation of astrocytes and toes scar formation can be reduced,and the spinal cord injury recovery of nerve conduction function in rats can be accelerated,for spinal cord injury rats the effect of combination was better than a single application.
