Back Grounds:In recent years, there are increasing trendency of spinal cord injury, its characteristics is hige incidence rate, high mutilation rate but low mortality. Spinal cord injury consists of primary injury and secondary injury. With the development of research, secondary injury including the imbalance of neurobiochemistry, microcirculation, etc, is one of the important causes result in permanenced nervous injury. Effective and prompt treatment can confine the lesion and promote functional recovery of nerves. Neurotrophin-3/neurotrophic factor 3, is the important member of neurothphin family, which can prevent embryonic and postnatal motoneuron death, maintain neuron survival, promote reparation and regeneration after nervous injury, regulate synapse activity of neuron. Experiment in vitro indicate NT-3 has satisfactory effect on neuronal survive and axonal growth, however, the effect research in vivo is less comparatively.Objective:To investigate the effect and the mechanism of NT-3 on reparation in spinal cord injury of rats through using the method of Allen's WD to make the animal model of acute spinal cord injured of rats, administrating NT-3 in subarachniod space, observing the change of motion evoked potential and hind limb motor function, detecting the level of SOD, NOS, MDA in blood and the express of apoptosis associated protein of nerve cell in spinal cord.Materials and Methods:Using the method of Allen's WD to make the animal model of acute spinal cord injured of rats, 105 healthy adult SD rats of either sex were randomly divided into three groups, control group , experimental group and sham group. A thin plastic tube was situated in subarachnoid space below the injured level for perfusion. Rats in experimental group received NT-3 20μl(including NT-3 200ng) from the tube at 0h,4h,8h,12h,24h and 3d,7d after injury .The saline-treated animals received the equal volume of normal saline at the same time as control.
