Objective:?1?To observe the changes of endogenous hydrogen sulfide in myocardial tissue of rats with cirrhotic cardiomyopathy at different stages, so as to investigate the value of H2S on pathogenesis of cirrhotic cardiomyopathy.?2?To observe the effect of different doses of Buyang Huanwu decoction on endogenous hydrogen sulfide in myocardial tissue of rats with cirrhotic cardiomyopathy, and to explore treatment method on cirrhotic cardiomyopathy.Methods:?1?40 SPF male SD rats, body mass 220250g, were randomly divided into blank control group?group N, n=9?, CCM model group?group M, n=31?.Group N was fed with ordinary diet and drunk tap water; First two weeks, group M was subcutaneous injection with CCI4?3ml/Kg, given double dose for the first time? two times a week combing with only drunk 10 percent ethanol, while 20 percent ethanol given for the next two weeks. From the fifth week the rat model with CCM start to drink30 percent ethanol. At the end of twelfth week, 9 rats?group M1?randomly selected from group M were killed, while the rest?group M2? were killed at the fourteen week. ALT?AST?ALB were detected. The concentration of H2S in the myocardium was measured. Myocardial and liver pathology were detected.?2?SPF grade SD male rats 70, weight about 220250g, according to the experimental design requirements, were randomly divided into control group?I group, n=10?, model group?n=60?. Rats in the model group were treated with carbon tetrachloride and ethanol to establish rats model with cirrhotic cardiomyopathy. After 12 weeks, the survival rats of model group were randomly divided into 4 groups: low dose group( ? group, n=11, given Buyang Huanwu Decoction 14g·kg-1·d-1 by intragastric administration)?middle dose group(? group, n=11, given Buyang Huanwu Decoction 21g·kg-1·d-1 by intragastric administration)and high dose group(?group, n=11, given Buyang Huanwu Decoction28g·kg-1·d-1 by intragastric administration), CCM model group(group ?,n=10, given normal saline 3ml·d-1 by intragastric administration),continue subcutaneous injection with CCI4, each time every other 3 days,300ml/L ethanol solution as the only drinking water, take food freely.The rats of control group were given normal saline 3ml·d-1 by intragastric administration. The treatment lasted for 8 weeks. The experiment lasted for 20 weeks; Serum level of c Tn I was detected. The content of H2S in the myocardium was measured; The liver and heart pathology were detected by HE staining.Results:?1??1? Liver function: the level of ALT and AST ascended, while ALB descend in the M1 and M2 group, it is significant differences with the N group?P<0.05?. The M2 group had significantly increase compared with M1 group, there are very significant differences?P <0.05?.?2?H2S in the myocardium: Compared with Group N, the concentration of H2S in the M1 group declined by up to 18 percent, it is significant differences with the N group?P <0.05?, the M2 group dropped 38 percent, there are very significant differences compared with group N?P <0.01?. The M2 group had greater decreases compared with M1 group, Which is very significant differences?P<0.05?.?3? liver pathology: N group were normal liver tissue, the M1 group and M2 group were hepatic cirrhosis, and M2 group developed severer compared with the M1 group.?4? Myocardial pathology: N group were normal myocardial tissue. Myocardial cell were normal in general terms but myocardium edema were seen in the M1 group. And we can observe myocardium edema and necrosis in the M2 group.?2??1?Change of Serum c Tn I levels: compared with control group, the level of serum c Tn I increased obviously in model group?group ???????? rats, and the difference was statistically significant?P<0.05?;Compared with CCM model group, the level of serum c Tn I in treatment group?group ?????? decreased, a statistically significant difference?P < 0.05?;?2?Content of H2S in the myocardium: compared with control group, the level of serum c Tn I has decreased in model group?group???????? rats, Which is very significant differences?P<0.05?;Compared with group ?, the level of serum c Tn I has statistically increased in treatment group?group ?????? rats;?3?Liver pathology:the group ? were normal liver tissue, the group ? were the most severe cirrhosis, group? were medium cirrhosis, group? were mild cirrhosis, and the severity of cirrhosis in group ? was more severer than group? and group?;?4?Myocardial pathology: group? were normal myocardial tissue, in group? there were myocardial necrosis,hypochromatosis, fibrosis and stripes pattern disappear. The myoplasm were irregular graininess.The myofiber were funicular. The inflammatory response was obviously. The degree of myocardial damage in group ?was obviously lighter than group ?. In group ? and group ? the size of myocyte necrosis was smaller than group ?,and with mild fibrosis and inflammation.Conclusions:?1?In the development of liver cirrhosis, the protection on myocardium get weakness with the decreased level of endogenous hydrogen sulfide,which may play a crucial role in the pathogenesis of liver cirrhotic cardiomyopathy.?2?Buyang Huanwu Decoction has the protective effect on myocytes on CCM, the mechanism may be associated with increasing the level of endogenous hydrogen sulfide.
