Objective Myocardial ischemia is the main cause of cardiovascular disease. The acupuncture therapy has the advantages such as less toxic side effect, convenient operation and low cost, meanwhile acupuncture has a good effect on the physiological activities of the heart, so it can effectively prevent and treat myocardial ischemia.The prevention and treatment effect of Biao-Ben acupoints compatibility method on chronic myocardial ischemia has analyzed the protective effect of Biao-Ben acupoints compatibility method on myocardial ischemia in rats, in this study, after the electro-acupuncture intervention treatment of chronic myocardial ischemia model rats, observing the change of enzyme activities of GADPH, CS and CCO in aerobic respiration, structure and function of myocardial mitochondria, S-T segments of ECG, morphology and behavior of rats.Methods In this experiment, 45 male Wistar rats, 3-month-old, weighting about 180~220g, were randomly divided into three groups: no-treatment control group(NC group), chronic myocardial ischemia model group(M group) and electro-acupuncture intervention group(EA group), with 15 rats in each group respectively. After 3 days of adaptive feeding, the rats in the NC group received injection of 0.9% sterile saline, 5mg/kg per day, for 7 days. Rats in the M and EA group received injection of isopropylarterenol hydrochloride, 5mg/kg per day, for 7 days to build the model of chronic myocardial ischemia. The rats of the EA group received electro-acupuncture intervention treatment from the 4th day to the end during this experiment. According to the Biao-Ben acupoints compatibility method, we choose ST36(Zu San Li)?RN4(Guan Yuan) and PC6(Nei Guan) in this study. Ipsilateral ST36 and PC6 composed a pair of electrodes, a shallow needling near RN4, about 5mm, composed another pair of electrodes with RN4.These electrodes connected with the Hans acupoint nerve stimulator(HANS-200) EA apparatus, selecting Intermittent wave at 2Hz-100 Hz in 1m, and stimulating 10 minutes per day for 21 days. Recording rats' S-T segments and the general situation at the 3rd day?10th days?17th days and 24 th day of the research. All the rats were taken tissue on the 25 th day, observing the grain ultra structure of cardiac tissue, testing the respiratory control ratio of mitochondrial, gauging the Content of ATP and the enzyme activities of GADPH, CS and CCO in myocardial tissue.Results 1. Comparison of the general situation of rats: The rats in the chronic myocardial ischemia model group had the following characteristics such as having secretions at canthus, Cyanosis mouth and tail, thinning hair, listlessness and reduction in diet. While the situation of the rats in electro-acupuncture intervention group was basically sound except some of them were easy to be frightened and to have narrow eyes. 2. Comparison of S-T segments of electrocardiographic examination: After modeling, the S-T segments of the M group and the EA group were significantly increased(P<0.01). The S-T segments of the EA group were recovered after electro-acupuncture intervention, and there was significant difference between the M group and the EA group(P<0.01). 3. Comparison of pathological structure of cardiac tissue: In the M group, the structure of myocardial fiber were destroyed and in disorder, myofibrils fracture, deformation intercalated disc and cytoplasm's stain is dark, karyopyknosis, interstitial myocardial edema and inflammatory cell infiltration, some pyknosis mitochondria, swelling or vacuolar lesions and mitochondrial cristae were broken into floc were observed under the optical microscope and TEM. While in the EA group, the structure of cardiac tissue was normal except for a few mild edemata among cells, interstitial and mitochondria. 4. Comparison of the contents of ATP in cardiac tissue: In the M and EA group, the contents of ATP in cardiac tissue were lower than that of the NC group, while the contents of ATP of the EA group were much higher than that of the M group, those differences were significant(P < 0.01). 5. Comparison of the RCRs of mitochondria: The RCRs of mitochondriain in the M group were reduced significantly at an abnormal level. The RCRs of mitochondria in the EA group were in the normal level and significant higher than that in the M group(P<0.01), but lower than NC group(P<0.01). 6. Comparison the activities of GAPDH in cardiac tissue: In the M and EA group, the activities of GAPDH in cardiac tissue were lower than that of the NC group, while the activities of GAPDH in the EA group were much higher than that of the M group, those differences were significant(P < 0.01). 7. Comparison the activities of CS in cardiac tissue: In the M and EA group, the activities of CS in cardiac tissue were lower than that of the NC group, while the activities of CS in the EA group were much higher than that of the M group, those differences were significant(P < 0.01). 8. Comparison the activities of CCO in mitochondria of cardiac tissue: In the M and EA group, the activities of CCO were lower than that of the NC group, while the activities of CCO in the EA group were much higher than that of the M group, those differences were significant(P < 0.01). 9. Analysis of the correlation: There are linear correlations between the RCR of mitochondria, the contents of ATP and the activities of GADPH, CS and CCO in cardiac tissue(P < 0.01). The R2 of Correlations were 0.886, 0.942, 0.908 and 0.949Conclusion 1. The model of chronic myocardial ischemia could establish by injection of isopropylarterenol hydrochloride successfully and conveniently. 2. The electro acupuncture intervention therapy of Biao-Ben acupoints compatibility could repair the damage causing by ischemia in cardiac tissue and play a positive role in the protection of structure and function of mitochondria by promoting the electrophysiological activities of the heart and improving the morphological expression of myocardium. 3. The electro acupuncture intervention therapy of Biao-Ben acupoints compatibility could increase the energy supply to the heart tissue so as to protect the cardiac tissue via activating the enzymes related to aerobic respiration and rising up the content of ATP in cardiac tissue.
