Objective:The correlation between acupoints and viscera is the quintessence of TCM Meridian theory.It is the earliest theory about somatic visceral interaction and has attracted much attention from scholars both at home and abroad.In recent years,the research on the correlation of meridians and viscera is mostly concentrated on the reflection(reaction)of the body surface meridian points after the body’s physiological function or pathological changes,and the target regulation effect on the physiological function or pathological changes of the viscera by the intervention of the meridians.Most of the observation results are observed,but the regularity is less summarized.There are many studies on the effect of acupuncture on the prevention and treatment of ischemic myocardial disease.The exact effect of Neiguan point on the prevention and treatment of myocardial ischemia is verified by multi-level and multi angle,but the mechanism of its effect is still not clear.Electroacupuncture is widely used in diseases of various systems,and its curative effect is affirmed.Frequency is one of the important parameters of electroacupuncture stimulation.It is generally believed that different electroacupuncture frequencies have different effects.At present,different electroacupuncture frequencies have not yet been recognized in the application of analgesia,but most of the studies support the protective effect of low frequency electroacupuncture on myocardial ischemic injury.Skin is one of the largest organs of the human body.The amount of skin’s blood perfusion is closely related to the state of the body,reflecting the blood circulation of the viscera.In the previous study,we found that the blood flow perfusion in the Neiguan(PC6)area can reflect the current state of the myocardium and its changes.According to the current research,sympathetic fibers in the heart are dominant and widely distributed.The amount of cutaneous blood perfusion is also controlled by sympathetic nerve,and sympathetic vasoconstrictor nerve is the most abundant vascular nerve innervation of skin and blood vessels.During acute myocardial ischemia,the excitability of the vasoconstrictor system of the sympathetic norepinephrine was enhanced,and norepinephrine and neuropeptide Y were released,which resulted in a series of pathological changes in myocardial contractility and energy consumption.In this study,the rat model of myocardial ischemia injury was prepared by coronary artery ligation.The effect of low frequency electroacupuncture point or non point on the pathological myocardium of ischemic injury and the changes of the times of treatment were compared and the cumulative effect of low frequency electroacupuncture point on myocardial ischemia were investigated.When the myocardium was in different states,the content of sympathetic nerve related substances and its receptors in the skin tissues of the related ponit and non point were determined and the changes with the times of EA treatment were also observed.This paper attempts to elucidate the scientific connotation of meridian-viscera related to the changes of molecular responses to sympathetic nerve-related receptors in order to open up a new way for the theoretical study of meridian-viscera-viscera.Methods:100 healthy male Wistar rats were randomly divided into one-time treatment group and three-times treatment groups.Then each group was divided into 5 groups:normal group,sham-operation group,model group,acupoint group and non-acupoint group.Each group contained 10 rats.On the second day after successfully producing the model of rats with myocardial ischemia,both therapeutic groups received low-frequency EA treatment one time a day.After the treatment of low-frequency EA on the first day or the third day,the content of serum CK-MB was detected by colorimetry and the content of serum cTnT were detected by ELISA method.And the content of NE,α1-AR,and β2-AR in the skin tissues of acupoint’s area detected by ELISA method.Results:Compared with the normal group,the contents of cTnT and CK-MB in the model group were significantly increased(P<0.01).Compared with the model group,the contents of cTnT and ck-mb were significantly decreased in the rats in the acupuncture group(P<0.01),and no significant change in the non-acupoint group(P>0.05).When the normal physiological state of myocardium is normal,the content of NE,α1-AR,and β2-AR in the Neiguan area is lower than that in the non-acupoint.Compared with the normal group,the content of NE and α1-AR in the model group’s Neiguan area was significantly increased(P<0.01),and the content of β2-AR decreased significantly(P<0.01).There was no significant change in the content of NE,α1-AR and β2-AR.Compared with the model group,the content of NE and α1-AR in the acupoint group was significantly decreased(P<0.01),and the content of β2-AR increased significantly(P<0.01).There was no significant change in the non-acupoint group(P>0.05).There was no significant difference in the content of NE,α1-AR and β2-AR comparison between the one-time group and the three-times group.Conclusion:(1)The low frequency EA therapy has protective effect on myocardial ischemia.(2)The sympathetic neuro-related substances in the skin tissues of the NeiGuan(PC6)area have the characteristics that the content of NE,α1-AR and β2-AR in skin tissue of Neiguan area varies with the different state of myocardium,and has a tendency to change with the different state of myocardium in rats.(3)The frequency of low-frequency EA has a certain influence on the the effect of acupoint.
