| ObjectiveTo investigate the expression of CXCR7 in human lung adenocarcinoma tissues and the correlation with Ki67 as well as their clinicopathologic significance. Besides, discuss the effect of abnormal expression of CXCR7 in the occurrence and the development of the lung adenocarcinoma.MethodsCollection of Affiliated Hospital of Nantong University in chest surgery 4 fresh lung adenocarcinoma tissues and corresponding normal lung tissues adjacent to carcinoma. Western Blot was used to detect the expression of CXCR7 in lung adenocarcinoma tissues and the normal lung tissues adjacent to carcinoma. We analyzed and collected the clinical data of 140 cases of lung adenocarcinoma and clinical pathologic characters. The specimens of the lung adenocarcinoma tissues and the normal lung tissues adjacent to carcinoma were made into tissue microarrays. Envision immunohistochemical staining was used to examine the expressions of CXCR7 and Ki67 in 140 cases of human lung adenocarcinoma specimens and 140 normal lung tissues adjacent to carcinoma. The relationship between the expression of CXCR7 and Ki67 in the pathological grade in human lung adenocarcinoma and clinical pathologic characters were analyzed by statistical methods.Results1. The Western Blot results suggested that: the expression of CXCR7 in lung adenocarcinoma tissues was significantly higher compared with the normal lung tissues adjacent to carcinoma.2. The IHC results showed that: the positive rate of CXCR7 in lung adenocarcinoma tissues(60.7%) was higher than that in the normal lung tissues adjacent to carcinoma(11.4%). Analysis with clinical data showed that: the expression of CXCR7 was closely related with the degree of differentiation(χ2=13.344, P<0.01), TNM staging(χ2=19.762, P<0.01), lymphnode metastasis(χ2=13.020, P<0.01), survival time(χ2=17.562, P<0.01). Besides, in the new pathological classification of lung adenocarcinoma cases, CXCR7 positive rate of the SPA(81.8%), MPA(66.7%) and the PPA(69.2%) was higher than LPA(36.4%) and APA(47.4%), the expression of CXCR7 was significantly different(P<0.01).3. The correlation analysis showed that: the CXCR7 expression was positively correlated with Ki67 in lung adenocarcinoma tissues(Spearman’s r = 0.705, P<0.01).4. Survival analysis showed that: By using the Kaplan-Meier analysis, patients’ middle survival rate with the positive expression of CXCR7(36.5%) was significantly lower than the patients with the negative expression of CXCR7(72.7%)(P<0.01); Moreover, the positive expression of CXCR7 and Ki67, whose survival rate was lower than the others(P<0.01).Conclusion1. The expression of CXCR7 in lung adenocarcinoma tissues was higher than that in the normal lung tissues adjacent to carcinoma; the expression of CXCR7 was closely related with the degree of differentiation, new pathological classification, TNM staging, lymph node metastasis.2. The expression of CXCR7 in lung adenocarcinoma tissues was positively correlated with the expression of Ki67. And the expression level was also different in new pathological classification of cases. The SPA, MPA, PPA were higher than LPA and APA. The cases with lower degree of differentiation and lymph node metastasis, the expression of CXCR7 and Ki67 were higher than the others. It indicated that CXCR7 and Ki67 might play important roles in the occurrence and development of lung adenocarcinoma.3. The patients with CXCR7 and Ki67 expression positive had a poor prognosis, and the survival rate was lower than the other patients(P<0.01). The new pathological classification, by detecting CXCR7 and Ki67 protein expression levels, might be helpful in the early diagnosis and prognosis evaluation of lung adenocarcinoma. |
