Objective:By building behavior evaluation, analyzing spinal cord tissue pathology, measuring spinal cord water content, evaluating blood-spinal cord barrier in rats after spinal cord injury. Observe the functional recovery and the influence of blood-spinal cord barrier of different concentration of rhubarb for spinal cord injury in rats, screen the best concentration of rhubarb and explore its mechanisms of action.Methods:Modified Allen's method is adopted to establish animal model of spinal cord injury. All 125 SD rats were divided into 5 groups randomly, they are control group, model group, rhubarb low group, rhubarb middle group and rhubarb high group, each group has 25 SD rats. Rats in control group were removed the lamina but no damage to the spinal cord. The control group and model group were given the same dose of normal saline (NS), and rats in each rhubarb groups were given rhubarb granules. In each time point keep a record of BBB score, measure spinal cord water content, evaluate blood-spinal cord barrier. After filling out then do HE staining. Observe the change of blood-spinal cord barrier after injecting evans blue by fluorescence microscope.Results:The experimental results show that, (1) BBB score of control group has no statistical difference at each time (P>0.05). Model group, rhubarb low, middle and high dose group have statistical difference at each time compared with control group (P<0.05). Rhubarb low dose group has no statistical difference at each time compared with model group (P>0.05). Rhubarb middle dose group has no statistical difference at 24h and 3d (P>0.05), has statistical difference at 7d and 14d (P<0.05) compared with control group. Rhubarb high dose group has no statistical difference at 24h,3d and 7d (P>0.05), has statistical difference at 14d (P<0.05) compared with control group. (2) HE staining:In control group, spinal cord tissue structure is clear, with no pathological changes. In model group the structure of spinal cord tissues is damaged, gray matter has flake bleeding while white matter is dotted hemorrhage stove, and syringomyelia is formed. In each rhubarb group, spinal cord tissue structure are all different degree damaged, bleeding from scattered flake and dotted hemorrhage, syringomyelia decreased significantly compared with model group. In rhubarb middle dose group, spinal cord tissue structure is damaged the lightest, spinal cord bleeding and syringomyelia are less than rhubarb low and high dose group. (3) Spinal cord tissue water content determination:model group, rhubarb low, middle and high dose group have statistical difference at each time compared with control group (P<0.05). Rhubarb low dose group has no statistical difference at each time compared with model group (P>0.05). Rhubarb middle dose group has no statistical difference at Oh (P>0.05), has statistical difference at 24h,3d and 7d (P<0.05) compared with control group. Rhubarb high dose group has no statistical difference at Oh,24h,3d (P>0.05), has statistical difference at 7d (P<0.05) compared with control group. (4) Fluorescence microscope finds that in control group there is no EB spilling, in model group there are lots of EB spilling particularly most in 3d group, the EB of each rhubarb groups have different degree of reduced compared with model group and EB spills the least in rhubarb middle dose group.Conclusion:(1) Rhubarb has protective effect on rats after spinal cord injury, it can effectively improve the recovery of rats double hind limbs motor function after spinal cord injury and the best group is the rhubarb middle dose group. (2) The mechanisms of rhubarb on spinal cord injury may be associated with protection of blood-spinal cord barrier, reducing the spinal cord bleeding, reducing the spinal cord tissue edema, improving local microcirculation damage area.
