| ObjectiveTo observe the clinical efficacy of chinese herbal compound Yangzheng Sanjie Decoction(YZSJD)in treating precancerous lesion of gastric cancer(PLGC),and to explore its mechanism of blocking precancerous lesion of gastric cancer through miRNA pathway.MethodClinical research:38 cases of chronic gastritis with intestinal metaplasia or/and dysplasia,including cases of spleen and stomach weakness type,spleen and stomach damp-heat type and liver and stomach discord type according to Syndrome Differentiation of TCM were collected.All patients were treated with modified Yangzheng Sanjie Decoction,and the changes of clinical symptoms and pathological conditions before and after treatment were observed.Experimental research:The N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)transformed eternalized human gastric mucosa epithelium GES-1 cell line(MC cell)was used in vitro as a model of PLGC.MC cells were cultured with 10%of YZSJD-containing rat serum(YZSJD group)or non-medicated rat serum(control group)respectively,then the cell proliferation was measured using Cell Counting Kit-8 assay,the rates of apoptotic cells were detected by flow cytometry of Annexin V-FITC/PI staining,and the expression of miRNAs was analyzed by real time PCR.Results1.After treatment with YZSJD,the clinical total effective rate was 97.37%.The histopathological examination results showed the total effective rate was 70%.The clinical symptoms and pathological conditions before and after treatment were significant different(P<0.05).2.In the YZSJD group,the cell proliferation was significantly lower than the control group(P<0.01),the proliferation inhibition rate was 10.67%.3.In the YZSJD group,the total apoptosis rate was 20.09±2.15%,the late stage apoptosis rate was 19.60 ± 2.19%;In the control group,the total apoptosis rate was 11.62±1.07%,the late stage apoptosis rate was 11.33±1.09%,,the difference was of statistically significant(P<0.01).4.The expression of let-7a and miR-7 in MC cells was obviously lower than that in GES-1 cells(P<0.05),but there was no statistical significant difference in the expression of miR-130a between the two groups(P>0.05).5.The expression of let-7a and miR-7 was up-regulated significantly in YZSJD group(P<0.01),while the expression of miR-130a showed no significant change(P>0.05).Conclusion1.Chinese herbal compound Yangzheng Sanjie Decoction has good therapeutic effects on blocking precancerous lesion of gastric cancer.2.Yangzheng Sanjie Decoction-containing serum can significantly inhibit proliferation and induce apoptosis in precancerous lesion of gastric cancer cells.3.Let-7a and miR-7 may act as tumor suppressors involved in the development of precancerous lesion of gastric cancer,and play an important role in the course of blocking precancerous lesion of gastric cancer by Yangzheng Sanjie Decoction. |
