| Objective:Through the observation of the effect of Liu Xi Decoction on serum liver function, hepatic fibrosis index, liver pathological changes and immunohistochemistry index of hepatic fibrosis rats were induced intraperitoneal injection of porcine serum. We aimed at researching the mechanism of Liu Xi Decoction treating hepatic fibrosis, to provide new ideas and methods of treatment for clinical treatment of hepatic fibrosis, to provide scientific theoretical basis for Liu Xi Decoction in clinical treatment of hepatic fibrosis.Methods:Selected 98 male Wistar rats, body weight around 95 grams and 105 grams. Feeding one week under normal conditions with enough food and water, randomly divided into seven groups:normal control group, model group one, model group two, herb prevention group, herb treatment group, western medicine prevention group, western medicine treatment group. The immunohepaticfibrosis model was induced by intraperitoneal injection of porcine serum. The normal control group and the two model groups were given 0.9% Sodium Chloride (0.02 mg/100 g) by intraperitoneal injection, two times a week for 7 weeks. Herb prevention group was given Liu Xi Decoction liquid (3 g/100 g). western medicine prevention group was given colchicines’oral liquid (0.02 mg/100 g). The model and the irrigation stomach, Modeling and orally at the same time,1 time a day for 35 days. Herb treatment group, western medicine treatment group began to administration at the 36th day. Measure the body weight once a week on a fixed time to adjust the dosage. At the 35th day, taking 7 normal control group rats, all the model group, prevention group rats, anesthetized rats by the way of ether inhalation. Blood was taken from the femoral artery, separate serum from the centrifuge, storage in-80 for refrigerator for the detection. Anatomy of rats to expose the liver. At the third mm from the edge of the left lobe of liver tissue and placed in formalin fixed and sections were stained with HE, Mallory staining and immunohistochemical staining, using an image analyzer, Quantitative and quantitative analysis of the chemical indicators of immunohistochemistry, the index of the data and the results of the statistical results. The weight of rats in each group was measured before each sampling. At the 70th day, The remaining rats were treatment according to the first method.Results:After the treatment of Liu Xi Decoction, hepatic fibrosis in rats serum liver function, hepatic fibrosis indexes and immunohistochemical indicators have been improved significantly, with the western medicine group compared with significant difference (P<0.01), The liver tissue pathological structure improvement in herb treatment group is better than western medicine group, immunohistochemical markers for the content was significantly lower expression was significantly inhibited, and western medicine treatment group compared with significant difference (P<0.01).Conclusion:Liu Xi Decoction can obviously improve the index of serum, protects hepatic tissue structure and function, inhibit the synthesis of collagen, inhibit expression of transforming growth factor beta 1, blocked the activation of hepatic stellate cells, and improving the energy metabolism of hepatic cells, stable hepatic cell structure and function, improve hepatic tissue blood micro circulation, protect the normal structure of hepatic lobule, inhibit the hyperplasia of fiber, prohibiting hepatic fibrosis progress. |
