From Traditional Chinese Medicine Dementia is a disease of abnormal mind because of the reduce of brain cells and brain dysfunction along with the aging,whose main performance is stupid,low intelligence, forgetful.The effect of Acupuncture treatment in AD is better than pure western medicine; the effect of Acupuncture combined with Traditional Chinese Medicine is superior to pure western medicine or Acupuncture alone. Acupuncture is an ancient treatment, has its own distinct theory, dredges the channels and collaterals,regulate Yin and Yang,reinforce the Healthy Qi and dispel the Evil Qi. Acupuncture in AD has broad space for development.Existing AD animal model can be summed up as genetically modified (gm) model and Non-gmo model. In Non-gmo models most only simulate a certain aspect of AD pathology characteristics,while the pathological process of AD is complex, various, and do not present the AD typical characteristic in the brain:senile plaques (SP) and neurofibrillary tangles (NFTs). Transgenic animal model is active and hot spot, it simulates the process of AD on the genetics, presents many AD typical pathology characteristics,such as Aβ deposition,SP and so on.Morris water maze is a classical procedure which is used evaluatad spatial learning and memory ability of animals,in the the research and application of neurobiology, neuropharmacology etc Morris has been widely used;in addition there are Diving platform experiment,Dark experiment,Shuttling box experiment,Eight arms maze,Y-electric maze, Lashley Ⅲ water maze. Each kind of experiment of learning, memory has its priorities and disadvantages,so we should be based on the characteristics of the experiment,our experimental needs,and experime-ntal conditions to choose suitable experimental installation.[objective] APP/PS1double transgenic AD mice is our research object, we acupuncture "baihui"and"yongquan"to discuss the AD model mice learning and memory and the cerebral cortex Aβ level, so as to provide a laboratory evidence for clinical treatment of AD.[Methods]16APP/PS1transgenic positive male mice were randomly divided into the model group and the electro-acupuncture group,8in each.At the same time there were8APP/PS1transgenic negative male mice in the normal control group.The electro-acupuncture group was acupunctured "baihui"and"yongquan",,then adopted the intervention of electrical needle equipment which was dilatational wave,15min at a time,every other day at a time,treatment lasted5weeks. Finally the ability of learning and memory of all mice was tested by Morris water maze.Using immunohistochemical imaging to study the levels of Cerebral Aβ in mice.[Results] Escape latency between three groups was statistically significant (P<0.05),escape latency between the model group and the electro-acupuncture group was statistically significant (P<0.05). Immunohistochemical results shown:Aβ1-40and Aβ1-42are mainly expressed in the nerve cells, which are mainly distributed in the place such as the cerebral cortex, hippocampus. In cerebral cortex the number of positive neuron cells of Aβ1-40is more than Aβ1-42. In Image pro Plus6.0analysis software we choose the integral optical density (I0D) to analysis Aβ.Aβ1-40and Aβ1-42of the model group express the highest, Aβ1-40and Aβ1-42of the electro-acupuncture group express lower than the model group.[conclusion] This experiment suggests electro-acupuncture can decrease levels of Aβ1-40and Aβ1-42in cerebral cortex of APP/PS1double transgenic mice,inhibit the production of Aβ.reduce the toxic effect of Aβ, and improve the state of AD.This experiment by Morris water maze prompt electro-acupuncture can improve learning and memory ability of the APP/PS1double transgenic mice; Through immunohistochemi-cal electro-acupuncture can reduce the level of Aβ1-40、Aβ1-42, we hypothesize that electro-acupuncture will probably improve learning and memory ability of APP/PS1double transgenic mice through reducing the level of Aβ.
