Objective Many researches confirms the close relation between the occurrence of endometriosis and such three pathological processes as adhesion—invasion—angiogenesis in endometrium. One of the three is occluded, which can prevent the occurrence and development of endometriosis. ICAM-1, MMP-2 and VEGF are the respective key factors of the three processes. On the basis of molecular and cell biology, the aim is to cultivate the respective endometrial cells for the control group, the eutopic endometrium group and the ectopic endometrium group. The fluorescence is used to make a quantifiable check of the influence of Shaofu(Lower Abdomen) Zhuyu(Stasisexpelling) Decoction on the mRNA expression of ICAM-1?MMP-2?VEGF in ectopic and eutopic endometrial cells of endometriosis. The further aim is to discuss the possible mechanism for the prevention and treatment of endometriosis and to provide the possible theoretical evidence for the clinical treatment of endometriosis by the application of the decoction.Methods (1)15 cases of patients with endometriosis in the type of cold coagulation and blood stasis were grouped into eutopic endometrium and ectopic endometrium, that is, eutopic endometrium group and ectopic endometrium group. Another 15 cases of patients with simple Uterine leiomyoma were classified as the control group. The grouping and classification is to cultivate the endometrial stromal cells for each group;(2)The real-time use of fluorescence was made to quantifiably check by means of PCR the mRNA expression of ICAM-1?MMP-2?VEGF in the third-generation endometrial stromal cells of each group.(3) Shaofu Zhuyu Decoction with the concentrations of 20g/L?10g/L?5g/L?2.5g/L?1.25g/L?0.625g/L?0.3125g/L were respectively added into the third-generation stromal cells in the eutopic endometrium group and ectopic endometrium group. The real-time use of fluorescence was made to quantifiably measure by means of PCR the after-adding-decoction mRNA expression of ICAM-1?MMP-2?VEGF in stromal cells of eutopic endometrium group and ectopic endometrium group. And the statistical analysis ensued.Results The result of the real-time use of fluorescence to quantifiably measure by means of PCR the endometrial stromal cells of each group showed that(1)the ICAM-1 mRNA expression of endometrial stromal cells in ectopic endometrium group was apparently higher than that in eutopic endometrium group and that in eutopic endometrium group was clearly higher than that in the control group;(2)the MMP-2 mRNA expression of endometrial stromal cells in ectopic endometrium group was apparently higher than that in eutopic endometrium group and that in eutopic endometrium group was clearly higher than that in the control group;(3) the VEGF mRNA expression of endometrial stromal cells in ectopic endometrium group was apparently higher than that in eutopic endometrium group and that in eutopic endometrium group was clearly higher than that in the control group; those differences were all of statistical significance(P<0.01). After the addition of Shaofu Zhuyu Decoction,the result of the real-time use of fluorescence to quantifiably measure by means of PCR the endometrial stromal cells of each group showed that(1) in the eutopic endometrium group, after the addition of the decoction, the ICAM-1,MMP-2 and VEGF mRNA expression was all lower than that in the group without adding the decoction;(2) in the ectopic endometrium group, after the addition of the decoction, the ICAM-1,MMP-2 and VEGF mRNA expression was also lower than that in the group without adding the decoction; those differences were all of statistical significance(P<0.01) and the lowering tendency was closely related with the concentration of the added decoction;(3)in the eutopic endometrium group, when the concentration of the decoction was rising to 2.5g/L, the ICAM-1,MMP-2 and VEGF mRNA expression reached the bottom. But with the increasing concentration of the decoction, the MMP-2 and VEGF mRNA expression began to show the rising tendency. However it rose slowly and the expression was kept lower than that in the group without adding the decoction;(4) in the ectopic endometrium group, when the concentration of the decoction was rising to 2.5g/L, the ICAM-1, MMP-2 and VEGF mRNA expression all reached the bottom. But with the increasing concentration of the decoction, the expression of the three factors began to rise. However it rose slowly and steadily and the expression was kept lower than that in the group without adding the decoction.Conclusion (1)The mRNA expressions of ICAM-1,MMP-2 and VEGF, the key factors in such three steps as adhesion-invasion-angiogenesis shows the striking differences in the endometrial stromal cells of the ectopic endometrium group, the eutopic endometrium group and the control group, which tells us that the occurrence and development of endometriosis is closely related to the three factors and that Shaofu Zhuyu Decoction can be regarded as an effective index for the prevention and treatment of endometriosis;(2) the prevention and treatment of endometriosis is likely to be related with the interference of such three programs as adhesion—invasion—angiogenesis, but which step is interfered needs to be further researched;(3)The application of Shaofu Zhuyu Decoction to the prevention and treatment of endometriosis is feasible to a certain extent. It can be tested by means of animal experiment and clinical experiment.
