BackgroundWound tissue repair is crucial for restoration of damaged tissue structure and function. Tourniquet, triangular bandage and gauze are first-aid methods which widely used to control damage currently. Although these traditional methods were constantly improved and derived various novel types, it still cannot meet the requirements of operation and trauma tissue repair. In order to improve self-help and mutual rescue during the war, natural disasters and other extreme conditions, liquid adhesive bandage emerge at the historic moment, a-cyanoacrylate compounds are the most widely used liquid adhesive bandage at present. Liquid adhesive bandage sealed and debride the small open wound quickly and convenient. However, the liquid adhesive bandage form a closed cavity after it treat wounds which creats a warm environment for bacterial growth. Improving the antibacterial property of the liquid adhesive bandage is imminent.5% three triclosan and 6% nano-silica were added to liquid adhesive bandage to improve the physical and chemical properties and improve the antibacterial effective have proved to be a good method, but it need to detect the antibacterial capacity and cytotoxicity of this novel liquid adhesive bandage.ObjectiveWe have proved the liquid adhesive bandage containing 5% three triclosan and 6% nano-silica possess strong antibacterial capacity in vitro. The purposes of these experiments were to evaluate the antibacterial properties and cytotoxicity of the novel liquid adhesive bandage through various animal and cell experiments.Methods(1) Measure the correlation between the intensity of light emission after double dilution detected by small animal imaging instrumentation and the units of bioluminescence S. aureus.(2) Establishing incision and excision animal models and infected with S. aureus. Treat the infected wounds with novel liquid adhesive bandage, ordinary liquid adhesive bandage and traditional method. Detection the changes of photon count of bacteria at different time with the help of bioluminescence imaging and compare the healing time and the thickness of the wound scar.(3) 1‰ novel liquid adhesive bandage were added into the culture solution and culture for 24h, then cell viability was determined by CCK-8 assay, the number of cells were detected by DAPI staining, cell apoptosis was detected by Annexin V-FITC/PI double labeling staining.Rusults(1) The luminous intensity of luminous S. aureus is highly relevant to the bacterial number.(2) The novel liquid adhesive bandage inhibit bacterial reproduction on the infected incision and excision wounds effectively.(3) The novel liquid adhesive bandage has low inhibition on the reproduction of mouse fibroblast L929 cell.(4) The novel liquid adhesive bandage can increase the volume of mouse fibroblast L929 cell.(5) The novel liquid adhesive bandage cannot induce mouse fibroblast L929 cell apoptosis.Conclusion(1) The novel liquid adhesive bandage has good antibacterial properties of bacteria infected wounds.(2) The cytotoxicity of novel liquid adhesive bandage has low cytotoxicigy, but it cannot induce cell apoptosis.
