| Objective:1. The study of angiotensin type 1 receptor(AT1R gene A1166 C polymorphism and region of Guizhou province, han the correlation of primary hypertension.2. To investigate the AT1 R gene A1166 C polymorphism and environmental factors interaction effect on primary hypertension.Methods:Selected302 cases of hypertension patients in trait region, Guizhou province(Buyi 154 cases,148 cases of Han nationality) for high blood pressure group, 299 cases of healthy subjects(Buyi 143 cases, 156 cases of Han nationality) as the normal control group,Using PCR amplification technology and restriction enzyme digestion method(PCR- RFLP) testing two national hypertension group and control group AT1 R gene A1166 C purpose AA, AC and CC genotype and A and C allele frequency distribution,and useing chi-square test, analysis of covariance, multivariate Logistic regression analysis AT1 R gene A1166 C polymorphism and the role of environmental factors on the pathogenesis of essential hypertension, as well as the gene polymorphism and environmental factors interact effect on primary hypertension disease.Results:1.(1) The Buyi people compared with the Han people, the genotype and allele frequency without significant difference;(2) AA, AC and CC genotype frequency in the hypertension group were 90.7%, 9.3% and 0, in the normal control group were 96.7%, 3.3% and 0, the more similar between the two groups was statistically significant(P=0.003).A and C allele frequency in hypertension group were 95.4%, 4.6%, and in the normal control group were 98.3%, 1.7%, are similar between the two groups have statistical significance(P=0.003);Hypertension group AC genotype and C allele frequency was significantly higher than that of normal control group.(3) AA, AC and CC genotype frequency in the Buyi hypertension group were 93.5%, 6.5% and 0, in normal control group and in the province were 95.8%,4.2% and 0, respectively, compared with similar between the two groups have no statistical significance(P=0.381);A and C allele frequency in the buyi hypertensiongroup were 96.8%, 3.2%, and in the normal control group were 97.9%, 2.1%, buyi is similar between the two groups has no statistical significance(P=0.388).(4) AA, AC and CC genotype frequency in the Han nationality hypertension group were 87.8%,12.2% and 0, while in the Han normal control group were 97.4%, 2.6% and 0,compared to similar between the two groups was statistically significant(P=0.001).A and C allele frequency in the Han nationality hypertension group were 93.9%, 93.9%,and in the normal control group were 98.7%, 1.3%, the Han nationality is similar between the two groups was statistically significant(P=0.002), the han nationality hypertension group AC genotype and C allele frequency was significantly increased.2.Interaction analysis shows that hypertension incidence of buyi people,AT1 R gene A1166 C polymorphism and gender, smoking, alcohol consumption, blood sugar interaction;Hypertension incidence of the Han people, AT1 R gene A1166 C polymorphism and drinking interaction exists.3. Multivariate Logistic regression analysis showed that multiple factors, adjust the gender, age, smoking, drinking, BMI,blood sugar, TC, TG, LDL-C and other risk factors, AT1 R gene A1166 C polymorphism and buyi people developing hypertension has no correlation(P=0.965),and hypertension morbidity associated with the han Chinese population;AC genotype population at risk of high blood pressure is 5.658 times of AA genotype groups(P=0.000).4. The covariance analysis showed that adjustment for age, BMI, blood glucose, TG, TC, LDL-C and other factors, AT1 R gene A1166 C polymorphism and buyi people no correlation between systolic and diastolic blood pressure(P=0.331, P= 0.987), and related to the Han nationality people in guizhou systolic blood pressure significantly(P=0.019), and its diastolic blood pressure with no correlation(P=0.786).5. Blood pressure classification and its influencing factors of orderly classification logistic regression analysis showed, buyi people blood pressure level increases with age, smoking status, alcohol consumption, higher BMI, blood sugar,TC,The han nationality population blood pressure levels and age, BMI, blood sugar,TC, TG, LDL- C increased, and AC genotype.Conclusion:1. AT1 R gene A1166 C polymorphism and trait of Guizhou buyi people in essential hypertension incidence,and primary hypertension disease related with the han Chinese population, AT1 R geneA1166C polymorphism may be independent risk factors for the development of the han population of primary hypertension.2. The Han people with AC, carrying C allele genotype increased risk for primary hypertension, AC genotype and C allele may be the han population of primary independent risk factors for high blood pressure and systolic blood pressure.3. AT1 R gene A1166 C polymorphism and gender, smoking,alcohol consumption, blood sugar of buyi people developing hypertension have interaction;AT1R gene A1166 C polymorphism and drinking to Han Chinese populations hypertension the interaction.4. Age, smoking, drinking, BMI, blood glucose, and increased the TC may be the independent risk factors of primary hypertension Buyi people;Age, BMI, blood sugar, TC, TG, LDL- C increased and AC genotype may be independent risk factors for the development of the Han population of primary hypertension. |
