ObjectiveThis study was designed to study renin-angiotensin-aldosterone hormone levelsand CYP11B2-344C/T gene polymorphism in essential hypertension patients withobesity, discussing genetic susceptibility of CYP11B2-344C/T genotype and thecorrelation between susceptibility gene and RAAS.Methods60cases of1to2grade essential hypertension patiens from Department ofCardiovascular Medicine, and60people from the Examination Center of SecondAffiliated Hospital of University of South China during2011–2012were enrolledrandomly. All subjects were divided into four groups:30cases essential hypertensionpatients with obesity (group A),30cases essential hypertension patients withoutobesity (group B),30cases obesity people without hypertension (group C),30caseshealthy people (group D). Patients were enrolled according to the hypertensiondiagnostic criteria of Hypertension Prevention Guide2010and diagnostic criteria ofthe prevention and control guidelines of Adult overweight and obesity of2004,excluding related diseases in accordance with the exclusive criteria. CYP11B2-344C/T gene polymorphisms were analyzed using Polymerase Chain Reaction-RestrictionFragment Length Polymorphism and agarose gel electrophoresis methods; PlasmaRAAS concentration were detected by radioimmunoassay; the data was statisticalanalyzed by SPSS13.0software.Results1.The comparison of RAAS hormone levels in each group Essential hypertension with obesity, essential hypertension, obesity group,plasma renin, angiotensin II and aldosterone levels were compared with health controlgroup, had significant difference (P<0.05).2.The comparison of CYP11B2-344C/T genotype and allele frequency distribution(1)The comparison of CYP11B2-344C/T genotype frequency distributionEssential hypertension with obesity group, essential hypertension and obesitygroup were multi-compared with number of cases of TT and TC+CC type genotype,having significant differences(x2were10.000,5.455,6.787respectively,P<0.05).Anddifferences in essential hypertension with obesity group was the most significant.(2) The comparison of CYP11B2-344C/T allele frequency distributionEssential hypertension with obesity group, essential hypertension group andpure obesity group were multi-compared with the number of T and C allele, havingsignificant differences (x2were13.297,7.548,10.159respectively, P<0.05). Anddifferences in essential hypertension with obesity group was the most significant.3.Relationship between CYP11B2-344C/T genotype and RAAS hormone level.According to Plasma Renin, Ang II, ALD concentration of genotype TT weremuch higher than TC and CC genotype,(P<0.05); comparing with CC,TC genotypehas no significant differences(P>0.05).Conclusions1.Renin-angiotensin-aldosterone hormone levels significantly increased, as comparedwith other groups, in essential hypertension with obesity group.2.In CYP11B2-344C/T gene polymorphism, T allele may be susceptibility gene ofessential hypertension patients with obesity,TT genotype may be susceptible genotypeof them.3.Each component of plasma renin-angiotensin-aldosterone hormone in the TTgenotype was significantly higher than the CC and the TC genotype,TT genotypemay be susceptible genotype of essential hypertension with obese patients.
