Objective:To observe the therapeutic effect of Yiqi Huayu(YQHY) recipe on dysfunctional uterine bleeding (DUB), and explore its possible mechanism to provide a scientific basis for clinical application.Methods:1. The male and female rats were fed in cages. Few days later, picking out the pregnant female rats and dividing them into six groups randomly. Groups were divided as follows:control group, model group, Yi-Fu bleeding pill group, YQHY recipe low-dose group, YQHY recipe middle-dose group and YQHY recipe large-dose group. On the 7th day, all groups except the control group were fed mifepristone and misoprostol as well, to established the dysfunctional uterine bleeding model in rats. Then, measuring hemorrhagic amount by placing cotton ball into vagina.2.50 female Kunming mice were randomly divided into five groups as follows: control group, Yi-Fu bleeding pill group, YQHY recipe low-dose group, YQHY recipe middle-dose group and YQHY recipe large-dose group. After intragastric administration, using capillary glass tube to method clotting time, evaluating the bleeding time by cutting tail.Female rats were randomly divided into five groups as follows:control group, model group, Yi-Fu bleeding pill group, YQHY recipe low-dose group, YQHY recipe middle-dose group and YQHY recipe large-dose group. Continuous lavage for 7 days. On the 6th day, giving warfarin 45 mg/kg to prepare model of blood coagulation dysfunction. After the last i.g., drew blood from abdominal aorta, preparing platelet rich plasma and platelet plasma respectively, measuring prothrombin time (PT), activated partial prothrombin time (APTT) and thrombin time (TT).3.60 SD female rats were assigned randomly into control group, model group, Yi-Fu bleeding pill group, YQHY recipe low-dose group, YQHY recipe middle-dose group and YQHY recipe large-dose group. On the 14th day, preparing blood stasis model by subcutaneous injecting of adrenaline hydrochloride and soaking in ice water. After the last i.g., collecting carotid artery blood, measuring platelet aggregation rate, deformation capacity, blood viscosity and erythrocyte aggregation and blood coagulation system indicators.4. Kunming mice, male and female, were randomly divided into control group, model group, Yi-Fu bleeding pill group, YQHY recipe low-dose group, YQHY recipe middle-dose group and YQHY recipe large-dose group. I.g. Senna immersion to prepare spleen deficient model. Injecting India ink, determinating meshy endodermis system to gobble up function. Picking up liver, spleen, weighing and calculating coefficient of devouring index K and a values.5. Picked up rats conceived for 7 days, i.g. mifepristone and misoprostol to copy model of dysfunctional uterine bleeding. Rats were randomly divided into control group, model group, Yi-Fu bleeding pill group, YQHY recipe low-dose group, YQHY recipe middle-dose group and YQHY recipe large-dose group. Prepared serum and uterus homogenate after gavage, detected estradiol (E2) and progesterone (P) in serum and endometrium by radioimmunity method.Results:1. Compared with the normal group, blood loss was significantly increased in the model group. The YQHY recipe could obviously reduce blood loss, compared with the model group(P<0.05, P<0.01). It indicates that dysfunctional uterine bleeding can be suppressed by YQHY recipe.2. Compared with the control group, bleeding and clotting time could be shorten notably in the high dose of YQHY recipe (P<0.05, P<0.01). But there was no significant difference between the low and medium doses of YQHY recipe when compared with control group in the decreasing of bleeding and clotting time(P>0.05). All of these show that high dose of YQHY recipe has some role in promoting clotting. TT, PT and APTT were shortened in warfarin-induced clotting disorders rats when using the medium and high doses of YQHY recipe (P<0.05, P<0.01). APTT is significantly shortened in the low dose of YQHY recipe (P<0.05). It shows that YQHY recipes has a good therapeutic effect on coagulation disorder.3. Compared with the model group, platelet aggregation could significantly decreased by using the doses of YQHY recipe (P<0.05, P<0.01). Red blood cell deformability could be strengthen in the medium and high dosages, erythrocyte aggregation could be decreased in the high dosage (P<0.05, P<0.01).Whole blood viscosity of the low shear rate (5s1, Is-1) could be decreased in the high dosage, Plasma viscosity can be decreased in both the medium and high dosages (P<0.05, P<0.01).6,12g/kg of YQHY recipe could decrease APTT. TT (P<0.05, P<0.01), and 12g/kg of YQHY recipe could increase FIB content (P<0.05), but all groups showed no effect on PT(P>0.05).4. Mice were fed with Senna immersion continuously.7 days later, great part of mice showed weight loss, loose stools, rectal prolapse, poor appetite, abdominal distension, spleen and other symptoms. Phagocytosis coefficient and phagocytic index were significantly increased under the medium and high doses of YQHY recipe (P<0.05, P<0.01), and the content of serum haemolysin was significantly strengthened, thymus and spleen index were increased in splenic asthenia mice (P<0.05, P<0.01).5. Compared with the model group, estrogen and progesterone levels had no significantly difference by using YQHY recipe (P<0.05). The estradiol level in endometria was notably increased (P<0.01) and progesterone level was notably decreased in model rats. YQHY recipe could decrease the level of estradiol and progesterone (P<0.05).Conclusion:There is a good therapeutic effect of YQHY recipe on dysfunctional uterine bleeding, it can significantly reduce the the amount of bleeding. The mechanism is related to promoting blood clotting, shortening the clotting time, improvig immune function, regulating endometrial local estrogen and progesterone levels.
