Objective:To observe the recovery of nerve function, human umbilical cord blood stem cells(hUCBSC) transplantation combined with EA therapy to spinal cord injury in rats.Methods:This experiment use the method of generation of centrifugal in primary culture to collect the umbilical cord blood stem cells, adjusting to a density of 1 × 107/ul,and preparing for Transplanting.72 female Sprague-Dawley (SD) rats, were randomly divided into 4 groups:1.the spinal cord injury(SCI) group; 2. human umbilical cord blood stem cell(hUCBSC) group; 3.Electronic-acupuncture (EA) therapy group; and 4. human umbilical cord blood stem cell (hUCBSC) combined with Electronic-acupuncture (EA) therapy (hUCBSC+EA) group. Each group had three time points:7days,14days and 28days. Making the spinal cord injury(SCI) models, (hUCBSC) group is distributed a cell transplant by Tail vein injection,which is 1.0×107/?L?(EA) therapy and (hUCBSC+EA) group are distributed electronic-acupuncture treatment once a day, The electronic-acupuncture time and frequency is set to 30 minutes dilatation wave 5 hz until taking the materials. The first, Recovery of nerve function in each group was assessed based on Basso-Beattie-Bresnahan (BBB) scores.The second, Using PowerLab data recording and analyzing system detects motion barrier conduction time evoked potentials; the last, taking the injured spinal cord by Electron microscope is to analyse spinal cord Morphological change.Results:1. BBB motor functional scores of the rat hind legs after spinal cord injury show, the joint group is different to the other three groups, and Limb function in rats can be quickly restored.2. Perspective Electron microscopic display,The myelin sheath arranged more regular in joint group, the cavitation of microtubules was decreased,and Lots of synapsis.the degree of swelling mitochondria was reduced. the thin regenerated myelin sheaths increased significantly.3. The detection of motor evoked potentials showed, the difference was insignificant between the EA and hUCBSC groups (P> 0.05).The joint group was different to the other three groups (P<0.05).Conclusion:The transplanted hUCBSC can survive, act on the injury after SCI. Electronic-acupuncture stimulation can promote the recovery of SCI; Umbilical cord blood stem cells combined with electronic-acupuncture were good at myelination in the nerve fibers, and the hindlimb moter functional recovery of SCI in rats.
