The Establishment Of Endometriosis Model In Allogeneic Sprague-Dawley Rats And The Expression Of MIF, MMP-9 In Rat With Endometriosis And Its Correlation | Posted on:2016-02-12 | Degree:Master | Type:Thesis | Country:China | Candidate:J X Cui | Full Text:PDF | GTID:2334330488999276 | Subject:Obstetrics and gynecology | Abstract/Summary: | PDF Full Text Request | Objective:To establish the allogeneic animal model of endometriosis (EMT) through the abdominal subcutaneous implantation, then to evaluate it.To investigate the expression of macrophage migration inhibitory factor (MIF) and matrix metalloproteinase-9 (MMP-9) in the rat endometriosis animal model and their relationship; Methods:Totally 50 health unmated adult female SD rats were selected. Monitor the estrous cycle of all the SD rats. They were randomly divided into donor and receptor group. Choose the SD rats which in the estrus cycle to establish the EMT model. The uterus of donor rat were sutured in the receptor’s abdominal subcutaneous. Heterotopic tissues and normal uterine tissues were taken for pathological examination on 3 weeks after surgery.Using the rat endometriosis animal model, to detect the expression level of MIF, MMP-9 in serum and lesions of normal rats and rats with endometriosis and their relationship by enzyme-linked immunosorbent assay (ELISA) and Immunohistochemical streptavidin-peroxidase (SP method).Results:(1) All experimental SD rats survived. The success rate of the allogeneic abdominal subcutaneous implantation was 66.67%. The histopathologic manifestations were similar to human endometriosis;(2) The expression of MIF and MMP-9 in the serum of rats with endometriosis were significantly higher than the normal rats, the difference was statistically significant (P<0.01); The expression of MIF and MMP-9 in the serum of rats with endometriosis and normal rats was positively related (r=0.389, P<0.05); (3) The expression of MIF and MMP-9 in the lesions of rats with endometriosis were significantly higher than the normal endometrium and eutopic endometrium in eutopic endometrium were higher than the normal endometrium, but the difference was no statistically significant (P> 0.05); The expression of MIF and MMP-9 The expression of MIF and MMP-9 in the serum of rats with endometriosis and normal rats was positively related (r=0.599, P<0.05)Conclusions:We successfully establish the rat allogeneic endometriosis model by abdominal subcutaneous implantation. It can be used as animal model in the endometriosis research.MIF and MMP-9 play an important role in the pathogenesis of endometriosis, and there is a relationship of mutual promotion between them. | Keywords/Search Tags: | Endometriosis, Animal model, Allogeneic, Subcutaneous transplantation, Macrophage migration inhibitory factor (MIF), Matrix metalloproteinase-9 (MMP-9), Rat | PDF Full Text Request | Related items |
