Objective:To explore the effect of Haobieyangyinruanjian on the mouse Liver fibrosis by schistosomiasis,and investigate the possible mechanisms in Therapeutic effect of liver fibrosis by effecting MMPs and TIMP-1?Methods:(1)Model Establishment:Mice were infected through skin of abdominal wall by 26(±1)Japanese schistosome cercarias,hepatic fibrosis was induced after 12 weeks.(2)Groups:The infected mice were randomly divided into 8 groups,namely Haobieyangyinruanjian low,middle and high dose treatment groups,positive control group(Fufangbiejiaruangan tablet control groups,Colchicine control groups),model contrast group(3months group positine infection,and 6 months group after infection),and normal group.Haobieyangyinruanjian groups and positive control group were given oral dose of the suspension,the model contrast group was given an equivalent dose of saline.once per day,within 12 weeks.(3)Items of Observation:Liver tissues and blood serum were obtained at the end of the experiment.To observe the pathological change of liver tissue under the microscope,to survey the contents of Hyp,the levels of HA and PC-III with RIA,the levels of MMP-2 and MMP-9 with the gelatin zymography,and the level of TIMP-1 by Immunocytochemical method.Results:(1)Histological examination and pathological grading:The degree of hepatic fibrosis in model contrast group(3months group positine infection,anrd 6 months group after infection)was significantly higher compared with normal contrast group(P<0.01),The degree of hepatic fibrosis in Haobieyangyinruanjian groups and positive control group was significantly lower compared with model contrast group(6 months group after infection).Haobieyangyinruanjian can reduce markedly the pathological changes.(2)The content of hydroxyproline in model contrast group(3months group positive infection,and 6 months group after infection)was significantly higher compared with normal contrast group(P<0.01),Haobieyangyinruanjian high,middle dose treatment groups were lower than model contrast group(6 months group after infection)(P<0.05).(3)The content of HA in model contrast group(3months group positine infection,and 6 months group after infection)was significantly higher compared with normal contrast group(P<0.01,P<0.05),Haobieyangyinruanjian middle,low dose treatment groups was lower compared with model contrast group(6 months group after infection)(P<0.05).(4)There were no significant differences among groups of the contents of PC?(P>0.05).(5)The level of MMP-2 and MMP-9 in model contrast group(3months group positive infection,and 6 months group after infection)was significantly higher compared with normal contrast group,Haobieyangyinruanjian high dose treatment groups were lower than model contrast group(6 months group after infection).(6)The level of TIMP-1 in model contrast group(3months group positine infection,and 6 months group after infection)was higher compared with normal contrast group,Haobieyangyinruanjian high,middle dose treatment groups were lower than model contrast group(6 months group after infection).Conclusion:(1)The model of schistosomiasis liver fibrosis in mice was successfully established.(2)Haobieyangyinruanjian has the therapeutic effect on the schistosomiasis liver fibrosis mice.3)one of the mechanisms of action may be to reduce the level of MMP-2?MMP-9 and TIMP-1,so that it can resistance the liver fibrosis.
