Purpose:To observe whether HGF can induce EMT in hepatic carcinoma HepG2 cell and breast cancer MDA-MB-231 cell,and how HGF affects the invasion and migration of the cells.And a preliminary study of its mechanismMethods:1.Morphological changes were observed by light microscopy.2.Using RT-PCR to analyse the gene expression of E-cadherin.Vimentin and NF-kB.3.The level of E-cadherin?Vimentin and NF-?B were detected by western blot.4.Transwell chamber was used to test the invasion and migration of the cells.5.The expression of protein E-cadherin?Vimentin and NF-kB on liver cancer and breast cancer was detected by immunohistochemical.Results:1.HGF could induce EMT in hepatic carcinoma HepG2 cell and breast cancer MDA-MB-231 cell.2.After HGF treatment,E-cadherin mRNA levels were significantly lower,Vimentin mRNA levels were significantly increased;E-cadherin protein levels were significantly reduced,Vimentin and NF-?B protein levels ware significantly increased.PDTC,one of the NF-rB inhibitors,could inhibit the change.3.The numbers of the invasion and migration of cells were increased(P<0.01).PDTC,one of the NF-kB inhibitors,could inhibit the increased invasion and migration of HGF-induced.4.In HCC,the expression of NF-kB was related to the degree of tumor differentiation,the tumor size and the level of AFP(P<0.05).The expression of NF-kB and the level of AFP were positively correlated(rs=0.317,P<0.05).In breast cancer,the expression of E-cadherin was associated with lymphatic metastasis(P<0.01),the expression of Vimentin was associated with the degree of tumor differentiation(P<0.05)and the expression of ER(P<0.01),the expression of NF-kB was associated with the tumor size.lymphatic metastasis(P<0.05)and the expression of ER(P<0.01).There was a negative relationship between the NF-kB and ER(rs=-0.295,P<0.01),a negative relationship between NF-kB and E-cadherin(rs=-0.350,P<0.01).Conclusions:1.HGF can induce EMT in hepatic carcinoma HepG2 cell and breast cancer MDA-MB-231 cell and make the invasion and migration of the cells increased,suggesting that EMT is closely associated with the development and metastasis of tumor.2.PDTC can inhibit the effect of HGF,suggesting that HGF may via NF-kB pathway mediate the occurrence of EMT and the invasion and migration of tumor cell.Different cell lines,different results.3.Tissue immunohistochemistry results of HCC and breast cancer showed that,there was a positive relationship between the NF-kB and AFP,a negative relationship between the NF-?B and ER,a negative relationship between NF-kB and E-cadherin.,suggesting that NF-kB plays an important role in tumor development and progression through the EMT process.
