Objective: The mi RNAs differentially expressed profile of serum between patients with Henoch-Sch?nlein purpura( HSP) and healthy Children were obtained by mi RNA chip technology. The target genes and the signal pathways regulated by those key mi RNAs were analyzed through bioinformatics. The role of mi RNAs in the development of HSP in Children are elucidated.Methods:Total RNA from the serum of 10 Children with HSP and 10 healthy Children are extracted.mi RNA chips are used to detect the differential expression profile of mi RNAs in serum between Children with HSP and healthy Children. The expression level of mi R-122 and mi R-149 are detected by q RT-PCR technology. And the targted genes and signal pathways regulated by the two key mi RNA are analysized by bioinformatics.Results:1. The differentially expression mi RNAs profile of serums between Children with HSP and healthy Children are obtained by gene chip. In Children with HSP, the number of significantly upregulated mi RNAs are 16, and the downregulated mi RNAs are 12.2. In Children with HSP, mi RNA with the most upregulated times is mi R-122, and have15.3413 times upregulation. mi RNA with the most downregulated times is mi R-149, and have-5.1245 times downregulation.3. In 10 Children with HSP, the mean values of mi R-122 are 172.8±7.2 and 31.2±5.5 in 10 healthy Children, the upregulated times are 5.5. In 10 Children with HSP, the mean values of mi R-149 are 18.4±1.6 and 65.7±3.5 in 10 healthy Children, the downregulated times are-3.6.The results of the gene chip and the q RT-PCR verification are consistent.4. There are 173 target genes regulated by mi R-122, and 445 target genes regulated by mi R-149.5. The signaling pathways involved in 173 mi R-122 regulated target genes include six pathways: Dicer, MAPKinase, FAS, AKT, ErbB, p53. The signal pathways involved in 440 mi R-155 regulated target genes include ten pathways: Cell cycle, Wnt, TGF-beta, p53, MAPK,Calcium, Apoptosis, AKT, Ras.Conclusion:1. mi RNAs differential expression profile of serum between Children with HSP and healthy Children are obtained. In Children with HSP, the number of significantly upregulated mi RNAs are 16, and the downregulated mi RNAs are 12.2. In Children with HSP, mi RNA with the most upregulated times is mi R-122, and mi RNA with the most downregulated times is mi R-149.3. The results of bioinformatics analysis show that there are many target genes and sinal pathways involved in mi R-122 and mi R-149 and their function are complex. They may have important function in the occurrence and development of HSP in Children.
