Chapter 1 The study of Relationship between mRNA Expressions of EZH2 with Invasiveness of Pituitary Adenomas.Objective: To detect the mRNA expression of EZH2 in invasive and non-invasive pituitary adenomas and explore the relationship of EZH2 mRNA in invasive pituitary adenomas.Methods: According to Hardy-Wilson and Knosp classification,27 specimens were divided into invasive group(17 cases) and non-invasive group(10 cases).QPCR was adopted to detect the mRNA expression of EZH2.The difference of EZH2 expression between invasive group and non-invasive group were analyzed.Results: EZH2 mRNA expression through QPCR in invasive pituitary adenomas was higher than non-invasive pituitary adenomas( t=-5.307,P=0.000).Conclusion: The expression of EZH2 mRNA was correlated with invasiveness of pituitary adenomas.Chapter 2 The study of Relationship between Bax mRNA and Bcl-2 mRNA Expression with the invasiveness of Pituitary AdenomasObjective: To detect the mRNA expression of Bax and Bcl-2 in invasive and non-invasive pituitary adenomas and to explore the relationship of Bax mRNA and Bcl-2 mRNA in invasive pituitary adenomas.Methods: According to Hardy-Wilson and Knosp classification,27 specimens were classified into invasive group(17 cases) and non-invasive group(10 cases).QPCR was adopted to detect the mRNA expression of Bax and Bcl-2.The difference of Bax mRNA and Bcl-2 mRNA expression in invasive group and non-invasive group were analyzed.Results: The results of q PCR showed that the expression of Bax mRNA did not differ between invasive pituitary adenomas and non-invasive pituitary adenomas(t=0.824,P=0.418).The Bcl-2 mRNA expression in invasive pituitary adenomas was higher than non-invasive pituitary adenomas(t=-3.602,P=0.001).Conclusion: The expression of Bcl-2 mRNA was correlated with invasiveness of pituitary adenomas,and the expression of Bax mRNA was not correlated with invasiveness of pituitary adenomas.Chapter 3: The Relationship between EZH2 mRNA?Bax mRNAand Bcl-2 mRNA Expression with invasiveness of Pituitary AdenomasObjective: To detect the mRNA expression of EZH2,Bax and Bcl-2 in pituitary adenomas and explore the relationship of EZH2 mRNA,Bax mRNA and Bcl-2 mRNA in pituitary adenomas.Methods: According to Hardy-Wilson and Knosp classification,27 specimens were classified into invasive group(17 cases) and non-invasive group(10 cases).QPCR was adopted to detect the mRNA expression of EZH2,Bax and Bcl-2.The difference of EZH2 mRNA,Bax mRNA and Bcl-2 mRNA expression in invasive group and non-invasive group were analyzed.Results: The results of q PCR showed that the mRNA expressions of EZH2 and Bcl-2 in invasive pituitary adenomas were higher than non-invasive pituitary adenomas(t=-5.307,P=0.000;t=-3.602,P=0.001).The Bax mRNA expression did not differ between invasive pituitary adenomas and non-invasive pituitary adenomas(t=0.824,P=0.418).Pearson correlation analysis indicated that EZH2 mRNA positively related with Bcl-2 mRNA(r=0.718,P=0.000),inversely related with Bax mRNA(r=-0.585,P=0.001),and Bcl-2 mRNA inversely related with Bax mRNA(r=-0.435,P=0.024).Conclusion: EZH2 mRNA and Bcl-2 mRNA co-promote invasive growth of pituitary adenoma.
