| Objective : We want to clear that the effect of “yi qi jie du fang” drug serum on proliferation, apoptosis of ACHN Rena Cancer Stem Cells.Methods:the use of high, medium, low “yi qi jie du fang”drug serum intervene ACHN Rena Cancer Stem Cells1. MTT assay will test that the effect of proliferation of ACHN Rena Cancer Stem Cells which were intervened by “yi qi jie du fang”drug serum2. Flow cytometry will be test that the effect of apoptosis of ACHN Rena Cancer Stem Cells which were intervened by “yi qi jie du fang”drug serum.3. Flow cytometry will be used To investigate the expression of P53 protein which were intervened by “yi qi jie du fang”drug serumResults:1. “yi qi jie du fang”drug serum can inhibit the proliferation of ACHN Rena Cancer Stem Cells. Proliferation rate of different groups were: Blank control group : 24 h:0.36%,48h:0.41%,72:0.43%, low chroma group: 24 h:15.7%,48h:32.7%,72h:44.3%, medium chroma group: 24h:36.7%,48h:53.3%,72h:63.7%, high chroma group: 53.3%,48h:67%,72h:77.7%. the difference was statistically significant(P <0.05).2. “yi qi jie du fang”drug serum can arouse the Apoptosis of ACHN Rena Cancer Stem Cells. Proliferation rate of different groups were: Blank control group:0.55±0.075%;low chroma group:3.48±0.505 %;medium chroma group:9.33±0.612%;high chroma group:18.36±1.511%. the difference was statistically significant(P <0.05).3. “yi qi jie du fang”drug serum can increase expression of P53 protein of ACHN Rena Cancer Stem Cells.the rate of P53 were: Blank control group:0.78±0.25%;low chroma group:7.24±0.80%;medium chroma group:11.72±1.19%;high chroma group: 17.68±1.06%. the difference was statistically significant(P <0.05).Conclusion:1.“yi qi jie du fang”drug serum can inhibit the proliferation of ACHN Rena Cancer Stem Cells.2.“yi qi jie du fang”drug serum can arouse the Apoptosis of ACHN Rena Cancer Stem Cells.3.“yi qi jie du fang”drug serum can increase expression of P53 protein of ACHN Rena Cancer Stem Cells. |
