Effects Of Serum Of Yiqi Jiedu Decoction On The Growth And Jagged1 Of ACHN Cell

Objective:ACHN human renal cancer cells were used to study the effect of Yiqi Jiedu decoction serum on proliferation,apoptosis and Jagged1 ligand in Notch signaling pathway of human renal cancer cell line ACHN under different concentrations and different time of action,and the effect of the serum of Yiqi Jiedu decoction on the proliferation and apoptosis of human renal carcinoma cell line ACHN was studied.Perfect the action mechanism of Yiqi jiedu recipe in treating renal cell carcinoma.Methods:(1)CCK8 method was used to observe the effect of high,middle and low concentration of Yiqi Jiedu decoction drug serum and blank serum on human renal cancer cell line ACHN for 24 hours,48 hours,and so on.Effects of 72 h on the growth inhibition of ACHN human renal carcinoma cells.(2)the apoptosis of ACHN cells was detected by flow cytometry after 24 h intervention with Yiqi Jiedu decoction serum.(3)RT-qPCR method was used to detect the apoptosis of human renal carcinoma cells.Effects of different concentrations of Yiqi Jiedu decoction on the relative expression of Jagged1 gene in ACHN cells after the same time.Results:(1)the serum of Yiqi Jiedu decoction could inhibit the proliferation of ACHN cells in a time and concentration dependent manner.(2)the serum of Yiqi Jiedu recipe couldpromote the apoptosis of ACHN cells in a concentration dependent manner.(3)Yiqi jiedu prescription could promote the apoptosis of ACHN cells in a concentration dependent manner.Fang could down-regulate the relative expression of Jagged1 gene in ACHN cells in a concentration-dependent manner.Conclusion:The serum of Yiqi Jiedu decoction can inhibit the proliferation of ACHN cells and induce apoptosis.The mechanism may be related to the down-regulation of Jagged1 ligand gene expression.
