Objective: To explore the possible role of ROMO1 in the process of intervertebral disc degeneration by detecting the expression of ROMO1 in nucleus pulposus tissue from different degree of degeneration of intervertebral disc and investigate the correlation of ROMO1 expression level, ROS content, the degree of intervertebral disc degeneration.Methods: The lumbar intervertebral disc nucleus pulposus tissues were collected from the patients who were performed spinal surgeries at the First affiliated Hospital of University of South China from June 2015 to December 2015..According to the Videman grading scoring criteria, the specimens were divided into normal control group(group A), mild degeneration group(group B),moderate degeneration group(group C), and severe degeneration group(group D).Each group include 10 samples. Immunohistochemical techniques was used to detect the expression of ROMO1 and fluorescent probes DCFH-DA was applied to test ROS levels in the nucleus pulposus tissue of degeneration disc. The Image proplus 6.0 softwared was employed to analysis the expression of ROMO1 and the SPSS18.0 statistical software was used for statistical analysis.Results: On the basis of immunohistochemical results, the mean option density of ROMO1 in the four groups of A, B, C and D were measured in order to be0.192±0.089, 0.328±0.048, 0.399±0.053, 0.468±0.098, respectively. The results of ROS detection were 132.961±15.149, 191.889±17.880, 218.056±12.845,243.501±30.279 separately in the groups of A, B, C, D. Analysis of variance and multiple comparisons between groups showed with the aggravation of intervertebral disc degeneration, the expression of ROMO1 and the content of ROS also increase gradually(p < 0.05). There was a positive correlation between ROMO1 expression level with the content of ROS in intervertebral disc(r =0.732, P < 0.05). There was also a positive correlation between ROMO1 expression level with the intervertebral disc degeneration degree(r = 0.603, P <0.05).Conclusion: 1. ROMO1 and ROS were highly expressed in the nucleus pulposus tissue of the degenerated intervertebral disc, which has a positive correlation;2.The expression level of ROMO1 was positively correlated with the degree of degeneration of intervertebral disc, which might be related to aggravated oxidative stress injury by increasing the content of ROS in the nucleus pulposus.
