Objective: Determination of the length of Telomere and the mRNA expression of TRF1, TRF2, POT1 Telomeres protection protein in peripheral blood mononuclear cells of Aplastic Anemia patients. In order to Explore the correlation of AA TCM dialectical type and peripheral blood mononuclear cells of the length of Telomere, and Telomeres protection protein TRF 1, TRF 2, POT 1 mRNA expression levels, Further investigate the essence of the AA from molec?lar and try to seek new treatment ideas.Method: 40 patients with AA were grouped in accordance with the TCM Syndrome Differentiation standard, "Shen yang xu group" 13 cases, "Shen yin xu group" 13 cases "Shen yin yang Bang xu group" 14 cases. And 20 normal human peripheral blood mononuclear cells were selected as control group. Using Real-time quantitative PCR method to determine the 60 cases of patients with the length of Telomere, Telomeres protection protein TRF 1, TRF 2, POT 1 mRNA expression levels in peripheral blood mononuclear cells. Observe the correlation of AA TCM dialectical type and peripheral blood mononuclear cells of the length of Telomere, Telomeres protection protein TRF 1, TRF 2, POT 1 mRNA expression levels.To study the correlation between Telomere, TRF 1, TRF 2 and POT 1 in peripheral blood mononuclear cells of patients with Aplastic Anemia.Res?lts: 1. Compare with the normal control group, the peripheral blood mononuclear cells of mRNA expression level of TRF 1, TRF 2, POT 1 of AA patients, there were significantly different(TRF 1, TRF 2, POT 1, P < 0.05). 2. The change of Telomere length in AA patients was significantly lower than that in the normal control group(P < 0.05). 3.Peripheral blood mononuclear cells of the expression levels of TRF l in AA patients of "Shen yin xu group", "Shen yang xu group" and "Shen yin yang liang xu group", were higher than that in normal control group significantly increased, were significantly different( "Shen yin xu group", "Shen yang xu group", "Shen yin yang liang xu group", P<0.01); Compare with the normal control group, the peripheral blood mononuclear cells of TRF 2 mRNA expression levels of "Shen yin xu group" and "Shen yang xu group" were higher( "Shen yin xu group", "Shen yang xu group", P<0.05); Compare with the normal control group, the peripheral blood mononuclear cells of POT 1 expression levels of "Shen yang xu group" is higher( "Shen yang xu group" P=0.039, P<0.05).4. AA patients of "Shen yang xu group" with Telomere length changes decreased compared with control group,( "Shen yang xu group" P=0. 019, P < 0.05).5.The mRNA expression level of TRF 2 in "Shen yin xu group" was elevated than "Shen yin yang liang xu group"( "Shen yin xu group" P=0. 034, P < 0.05).Conclusion: 1.TRF 1, TRF 2, POT 1, Telomeres may be involved in the pathogenesis of Aplastic Anemia, and the high expression of TRF 1, TRF 2, POT 1 may be associated with Telomere shortening are closely related. 2. Short Telomeres play an important role in Aplastic Anemia in the onset or progression of disease, that Telomere length in "Shen yang xu group" was the lowest, thus "Shen yang xu group" of Aplastic Anemia patients with Telomere length may be more obviously shortened, and Telomere loss is the most serious. TRF 1, TRF 2, POT 1 is may be related to the Telomere shortening process, and may with the prognosis of Aplastic Anemia. 3. Showed negative correlation, POT 1 and the expression of TRF 1 there was a positive correlation between Telomere length and TRF 1, TRF 2 expression, suggesting that TRF 1, TRF 2 is likely involved in the process of damage the Telomere shortening, POT 1 may through the influence of TRF 1 indirectly involved in the process, the incidence of short Telomeres and Aplastic Anemia are closely related.
