Objective: Determination of Chronic aplastic megaloblasts in peripheral blood andbonemarrow mononuclear cell the length of telomere, telomerase reverse transcriptase(hTERT) mRNA expression levels and human telomerase associated protein1(hTP1),In order to Explore the correlation of CAA TCM dialectical type andperipheral blood and bonemarrow mononuclear cells of the length of telomere, hTERTmRNA expression levels and human telomerase associated protein1(hTP1),Furtherinvestigate the the essence of the CAA from molecular and try to seek new treatmentideas.Method:39patients with CAA were grouped in accordance with the ChineseMedicine of the Standard,“Shen yang xu group”13cases,“Shen yin xu group”13cases,“Shen yin yang liang xu group”13cases, Determination of39cases of CAA in patientswith peripheral blood and bonemarrow mononuclear cells of the length of telomere,hTERT mRNA expression levels and human telomerase associated protein1(hTP1) usingReal-time quantitative PCR method,and selected30cases of normal physicalexamination as a control group. Observe the correlation of CAA TCM dialectical typeand peripheral blood and bonemarrow mononuclear cells of the length of telomere,hTERT mRNA expression levels and human telomerase associated protein1(hTP1);Observe the correlation of peripheral blood and bonemarrow mononuclear cells of the length of telomere, hTERT mRNA expression levels and human telomeraseassociated protein1(hTP1) and CAA in patient age, gender.Results:(1) Compare with the normal control group, the peripheral bloodmononuclear cells of the length of telomere of CAA patients, there is no significantdifference(p>0.05), the peripheral blood mononuclear cells of hTP1expression levels ofCAA patients is higher(p<0.05), the peripheral blood mononuclear cells of hTERTexpression levels of CAA patients is lower(p<0.05).(2) Peripheral blood mononuclearcells of the length of telomere、the expression levels of hTP1and hTERT of CAApatients in “Shen yin xu group”“Shen yang xu group”and “Shen yin yang liang xugroup”, between the two groups there are no significant differences(p>0.05); Comparewith the normal control group, the peripheral blood mononuclear cells of hTP1expression levels of “Shen yin yang liang xu group” is highe(rp<0.05); Compare withthe normal control group, the peripheral blood mononuclear cells of hTERT expressionlevels of “Shen yin yang liang xu group” is lower(p<0.05);(3) Group CAA patientsby age, Peripheral blood mononuclear cells of the length of telomere of CAA patients in>40-year-old group,≤40-year-old group, there are no significant differences(p>0.05);Peripheral blood mononuclear cells of hTP1expression levels of CAA patients>40-year-old group is higher than CAA patients≤40-year-old group(p<0.05);Peripheral blood mononuclear cells of hTERT expression levels of CAA patients>40-year-old group is lower than CAA patients≤40-year-old group(p<0.05);(4)Group CAA patients by sex, Peripheral blood mononuclear cells of the length oftelomere、the expression levels of hTP1and hTERT of CAA patients Male group, CAApatients female group, there are no significant differences(p>0.05);Conclusion: Peripheral blood mononuclear cells of hTP1expression levels of CAApatients in “Shen yin yang liang xu group”is high, Peripheral blood mononuclear cells ofhTERT mRNA expression levels of CAA patients in “Shen yin yang liang xu group”islow; the peripheral blood mononuclear cells of hTP1expression level of CAA patients>40-year-old group is high, the peripheral blood mononuclear cells of hTERT mRNAexpression level of CAA patients>40-year-old group is low, Show that Peripheral blood mononuclear cells hTERT mRNA expression level and telomerase activity reducedmay be one of a Molecular Biology Pathogenesis of CAA patients in “Shen yin yangliang xu group”and>40-year-old group. But the high expression of hTP1probably isreflected. Speculate that Peripheral blood mononuclear cells hTERT mRNAexpression level、hTP1expression level and telomerase activity reduced may be one ofthe the manifestations of Molecular Biology of “Shen xu”.
