Effects Of Jia Jian Yuan Hua Ling Gan Powder On GSH And TNF-? Of Acute Liver Injury Mice Induced By Acetaminophen

Objective:This topic expounds the protection mechanism of Jia Jian Yuan Hua Ling Gan Pulvis against acute liver injury in mice caused by what acetaminophen,through the observation of GSH and TNF-a.It provides a theoretical basis for the clinical application,so as to traditional Chinese medicine treatment of the disease to lay a solid foundation,through normally scientific animal experiment and the experience.Methods:Seventy SPF male mice adaptive feeding for 7 days,weighted 18-20g,were randomly divide into six groups for ten mice in each group including negative control group, model group, tiopronin control group, higher-, medium-and lower-dose of Supplemental Jia Jian Yuan Hua Ling Gan Pulvis treatment groups.Before establishing models, higher-, medium-and lower-dose groups were respectively perfused with the dosage of 24.4g/kg/d,12.2g/kg/d,6.10g/kg/d for successive 14 days.Positive group was administered by tiopronin with the dosage of 0.075g/kg/d.Simultaneously,control group and model group were given equal volume of normal saline. The mice were given a stomach filling at 10 am every day.Then all groups were perfused once to make model with the dosage of 500mg/kg of acetaminophen except for contol model.After 4 hours the levels of ALT,AST ALP,GSH,TNF-a and ALP in the Serum were measured and pathological morphology of the liver tissue was observed.Results:(1)Compared with control group,the level of ALT,AST and ALP in the serum of every group were obviously decreased(P<0.01),especially in higher-dosage of Jia Jian Yuan Hua Ling Gan Pulvis;(2)Compared with control group,the level of GSH in every group were obviously elevatory(P<0.01),especially in higher-dosage of Jia Jian Yuan Hua Ling Gan Pulvis;(3)Compared with control group,the level of TNF-a in every other group were obviously decreased(P<0.01),especially in higher-dosage of Jia Jian Yuan Hua Ling Gan Pulvis;(4)Compared with control group,each dosage of the group could ameliorate the injured liver cells in different level(P<0.05),higher-dosage of Jia Jian Yuan Hua Ling Gan Pulvis were the most effective one.Conclusion:Jia Jian Yuan Hua Ling Gan Pulvis can be against with mode of acute liver injury mice,and maybe improve GSH reduce TNF-? of mice.TNF-a can injure liver.The mechanism of Jia Jian Yuan Hua Ling Gan Pulvis protecting liver maybe improve GSH and reduce TNF-a.It can fight free radicals,improve inflammation,inhibit apoptosis,liver cell degeneration and necrosis is to be achieved.
