Effect Of Bu Qi Huo Xue Tong Mai Wan On The Quantity And Function Of EPCs In Peripheral Blood Of Patients With Arteriosclerosis Obliterans

Objective Through the acquisition of peripheral blood Endothelial progenitor cells(EPCs)in patients with Atherosclerosis Obliterans(ASO)in Chinese medicine Bu Qi Huo Xue Tong Mai Wan intervention,to observe the effect of Bu Qi Huo Xue Tong Mai Wan on the quantity and function of EPCs in patients with ASO,to explore the intervention of Bu Qi Huo Xue Tong Mai Wan on arteriosclerosis occlusive disease,and to study the therapeutic effect of Bu Qi Huo Xue Tong Mai Wan on ASO.Methods 1.Cultivation and identification of EPCs in peripheral blood of patients with ASO.Isolated mononuclear cells from peripheral blood of patients with EPCs by Ficoll-density contrifugation,The cells were seeded on fibronectin coated culture flask,DMEM culture medium with 4d,transfer liquid,culture to 6d,collect adherent cells,laser scanning confocal microscope and flow cytometry were used to identify vascular endothelial progenitor cells(EPCs).2.Effect of Bu Qi HuoXueTongMaiWan on the quantity and proliferation,migration and adhesion of EPCs.After cultured 6 days,randomly divide the EPCs of the adherent wall into four groups: control group and Bu Qi Huo Xue Tong Mai Wan 30g/L?60g/L?90g/L group.EBM-2 culture medium with blank serum,30g/L ? 60g/L ? 90g/L Bu Qi Huo Xue Tong Mai Wan containing serum containing 72 h.Use the inverted microscope to observe the growth condition of EPCs,Respectively,the number of cell counting,absorbance and Transwell chamber assay was used to observe the EPCs,value-added ability and migration ability and adhesion function.Results 1.Cultivation and identification of EPcs.Isolated mononuclear cells were cultured for up to sixth days to become spindle shaped endothelial cells.By laser confocal microscope,Dil.ac LDL and Fl TC.UEA-I double staining positive cells are differentiated EPCs,flow cytometry analysis,after sixth days of culture the expression of CD34,CDl33 and VEGFR-2 cells respectively(18.2 + 6.3)% and(28.5 + 7.2)% and(77 + 6.4)%.2.Effect of Bu Qi Huo Xue Tong Mai Wan on the quantity and function of EPCs in peripheral blood of patients with ASO.The results show,compared with the control group,30g/L,60g/L,90g/L Bu Qi Huo Xue Tong Mai Wan containing serum could significantly increase the number of EPCs,60g/L,Bu Qi Huo Xue Tong Mai Wan containing serum had the obvious effect on the quantity and function of EPCs.Conclusion Bu Qi Huo Xue Tong Mai Wan can significantly improve the quantity and function of EPCs in peripheral blood of patients with ASO,This may be one of the mechanisms for the prevention and treatment of arteriosclerosis occlusive disease.
