Objective:(1). To investigate the effect of PUM2 gene over-expression on the proliferation and invasion abilities of human colon cancer SW620 cells.Methods:SW620 cells were transfected with plasmid pc DNA3.1(-) / PUM2(PUM2 group)or pc DNA3.1(-)( vector group). The m RNA and protein levels of PUM2, were evaluated by real-time PCR and Western blot, respectively. The effects of PUM2over-expression on the proliferation,invasion and apoptotic abilities of the SW620 cells were evaluated by CCK-8 assay, Transwell test and Flow Cytometryrespectively.Results:The PUM2 expression level in SW620 cells in PUM2 group was significantly higher than that in vector group(P < 0.05), the roliferation, invasion and apoptotic abilities of SW620 cells in PUM2 groups decreased significantly as compared with those in vector group(P<0.05).Conclusion:The over-expression of PUM2 significantly inhibits the proliferation, invasion and apoptotic of SW620 cells. |
