Provide animal experiments basis for patients who with fracture of jaw bone healing for long-term drinking carbonated drinksMethods Sixty 2-month--old female SD rats were randomly divided into two groups :experimental group( selcect commericial packaging of Coca-Cola),the control group(drinking water), 30 rats in each group. The study was performed after the rats were allowed to acclimate for 2 weeks, in the experimental group,began to fed Coca-Cola,average only 40 ml, after 12 weeks, measurement the bone mineral density of two groups. Preparation of rat model of mandibular fractures in the edge of the mandibular body of rats. Make a uncomplete mandible fracture through the buccal and lingual side at the size of one-millimeter-wide and three- millimeter-length at the end of maeteric crest of the rats. When 2 weeks, 4 weeks, 6 weeks, 8 weeks after surgery, respectively take the manible specimen of the rats from two groups. And then to detect the doublt X-ray bone density and march the X-ray detection. After that, the specimens through decalcified,tissue paraffin-embedding and serial sectioning, HE stainning,and to observed parallel BMP-2,IL-1 immunohistochemical detection. Use SPSS19.0 statistical software analysis the data of two groups are or have statistical significance..Results 1 Fed Coca-Cola to experimental rats, after 12 weeks, Comparison of bone mineral density with control group rats the proximal tibia,significant differences(P<0.05). 2 Studied by X-ray radiography in combination and bone mineral density on fracture-healing: the X-ray can rough reaction fracture healing of rats. At same time point, in experimental group fracture healing were slower than control group. After fracture 2 weeks, in two groups are all can visible a few callus formation which in the fracture line near the distal subperiosteal.fracture line was still clearly legible; According to the statistical analysis.no significance.(P>0.05). At 4 weeks, the most specimens has appear discontinuous callus which in the experimental group. At 6 weeks, the control group had reached standard of osseous healing,the fracture line has changed fuzzy which in the experimental group.At 8 weeks, two groups had reached standards of osseous healing. 3 The results of BMD test at new bone fracture callus : Post operatioin of mandibular fracture, at different time points, The new generated bone callus of rats in experimental group is lower than the normal control group(P<0.05), difference was statistically significant. 4 Observed by HE staining: Post operatioin of mandibular fracture two weeks, The experimental group compared with control group, less newgenerated bone callus on fracture site, bone trabecular spacing wider, bone trabecular narrower. Post operatioin of mandibular fracture four weeks, the new bone trabecular has long and thin,and arranged disorderly, spacing increase. A large number of osteoclast has appeared; Post operatioin of mandibular fracture six weeks, new bone trabecular arrangement is still mixed and disorderly. Seperation distance continue increase(P<0.05).Post operatioin of mandibular fracture eight weeks, the fracture callus formation in experimental were less than the control group.and bone trabecular arrangement even more mixed and disorderly, wide space, At around can visible the osteoclasts and the spindle of osteblast( P<0.05). 5 The results of BMP-2 test in callus tissue: Post operatioin of mandibular fracture two weeks, both in two group can visible a large number of cells expression was strongly positive. Post operatioin of mandibular fracture four weeks, the positive cells express clearly in experimental group. But the number of positive expression were less than 2wk, Post operatioin of mandibular fracture six weeks,can see a few positive cells in two group. Post operatioin of mandibular fracture eight weeks, casual to see positive cells in experiment group.Post operative at 2 w, 4 w, 6w, experimental group and control group compared the MOD value, Significant difference was statistically significant(P<0.05). 6 The results of IL-1 test in callus tissue:Post operatioin of mandibular fracture two weeks, both in two group can visible a large number of cells expression positive, Osteoclasts is positive expression cells;At 4 weeks,the IL-1 expession in control group change weaken.At 6 weeks, the IL-1 expession in control group are obviously weakening. No difference between the two groups. At 8weeks, the IL-1 expession in control group probably change negative expression. Part of the osteoblast showed positive expression in experiment group. Measurement two sets of IL-1 MOD values of positive expression.increased at first and then decreased.Dealt with statistics, The values of MOD are statistically significant.Conclusions 1 Animal experiments show that the rats has Long-term consumption carbonated drinks, after 12 weeks it bone mineral density change lower, bone trabecular become thinner, Osteoporosis model was established successfully. 2 Animal experiments show that the fracture healing of rats compared with normal control group which has long-term consumption carbond drinks, 3 long-term consumption carbonated drink can inhibition the positive expression of BMP-2,and to promote the positive expression of IL-1 in rat fracture healing area. |
