ObjectiveTo explore the efficacy of dipsacus bone mixture in bone fracture healing, its effects on blood biochemical indicators, its mechanisms on regulating nitric oxide synthase(NOS) expression, and its effects on calcitonin gene-related peptide (CGRP) expression.Methods1.The bone fracture models were established by making 3mm×3mm defects in the right tibia bones in SD rats. Then the 64 SD rats were divided randomly into two groups:control group and treatment group. Intragastric administration of dipsacus bone mixture concentrated solution were given to the treatment group.2.The following signs were observed 4d,7d,14d,28d after operation:①The formation of callus was monitored by X-ray in fracture specimens,②Blood biochemical parameters were detected in blood samples of rats,③The expression of NOS was detected by western blot in fresh callus samples of rats,④The expression of CGRP was detected by HE staining and immunohistochemistry in callus samples of rats.Results 1. Serum ALP were significantly increased on 7d,14d,28d (P<0.05 or P<0.01) which compared with control. Serum calcium and phosphorus ion concentration products were significantly raised up (K<0.01) on 4d,7d,14d.2. The expression of eNOS was significantly increased on 4d and 7d after operation (P<0.05) which compared with control. The expression of iNOS was also significantly increased (P<0.05 or P<0.01) during 7~28 days after operation.3. The CGRP expression was significantly increased on 4d, 7d,14d (P<0.05 or P<0.01) which compared with control. Fracture appearance of specimens and X-ray evaluation:4. X-ray evaluation of the control group and treatment group in 4th days after fracture were poor. There were no callus coverage, and bone defect was still significantly in the surgical. Visible under the naked eye after dissection of the hematoma, a large number of machines, was jelly-like.7 days after the fracture callus coverage, bone density increased slightly at the shadow, the control group with no significant difference between the treatment group.14 days after the fracture compared with the previous film has the density increased, and there were some differences in the control group rate of 62.5% excellent, excellent rate of 87.5% in treatment group.28 days after fracture, two group of bone defect occurred under both X-high density, some have abnormal healing, no obvious difference.Conclusion1.The pharmacological mechanism of Dipsacus mixture in bone fracture healing might be to increase the activity of serum ALP, increase calcium and phosphorus product, promote the calcium deposition, thereby promote fracture healing.2.Dipsacus bone mixture promotes fracture healing during 2 weeks after operation by mechanisms involving its regulations on the expression of CGRP and the activity of NOS.3. The experiment supplied a visual and objective provement for The Effects and its Mechanism of Dipsacus Bone Mixture in Bone Fracture Healing with Bone Fracture Models in the Right Tibia Bones in SD Rats. It enlarges a train of thought for treat Bone Fracture Healing.
