Objectives To explore protective effect of Low-molecular-weight Heparin Calcium(LHC) combined with Trimetazidine on intestinal acute mesangial vein thrombosis(AMVT) model of rats in the intestinal smooth muscle and mechanism of effect.Methods 1. The experimental group and drug intervention: A total number of 160 SD male rats were divided into four groups randomly, each group 40 rats,LHC+Trimetazidine group(group LT), LHC group(group L), Trimetazidine group(group T), Normal saline group(group N). To establishing a stable model of superior mesenteric venous thrombosis by blocking superior mesenteric vein of 8cm and the edge vein arch. 2Each group of drug intervention: 1)Normal saline group(group N): After the AMVT model establishing,rats were skin-poped normal saline(0.2ml/100g) per 12 h until 72 h after surgery.2)Trimetazidine group(group T): After the AMVT model establishing, rats were Trimetazidine(10mg/Kg) tailed vein injection per 12 h until 72h(0.2ml/100g) per 12 h until72h after surgery. 3)LHC group(group L) : After the AMVT model establishing, rats were skin-poped LHC(30U/100g) per 12 h until 72 h after surgery. 4)LHC+Trimetazidine group(group LT): After the AMVT model establishing, rats were skin-poped LHC(30U/100g) and Trimetazidine(10mg/Kg) tailded vein injection per 12 h until 72 h.3Observing general celiac appearance, Vena cava blood samples and intestinal segments were collected sequentially at 6h ?12h? 24h? 48 h and 72 h afrer surgery each group.Intestinal tissue were taken from the rats for inestinal tissue section, stained with hematoxylin and eosin, examined under light microscopy and evaluated histopathologically using Mesemeche scoring system at different time. The levels of MDA ? CK in the serum and the levels of ATP in the intestinal tissue samples were measured with ELISA. 4. The measurement data were expressed by`?ąs and analyzed by one-way ANOVA and the among groups of enumeration data were tested by LSDt.Statistics analysis using the SPSS 21.0 statistical program, P<0.05, the difffernce has statistical significance.Results 1.Observering rat abdominal cavity generally and The amples of intestinal wall were examined by light electron:Group N: Rats can eat food after operation, there was three rats died at 36 h afrer surgery. The majority of intestine had adhered and fund a small punch. there were large amounts of bloody ascites in abdominal cavity.Group T: Rats can eat food after operation, there was a rat died at 36 h afrer surgery, A few intestine had adhered.Group L: All of the rats were living and can eat food after operation, there were no intestinal adhereing.Group LT: All of the rats were living and can eat food after operation,there were no intestinal adhereing. 2Comparing with group N, Histopathology score of the rat intestine decreased at different time point in group LT?group L?group T(P<0.05),the histopathology score of the rat intestine in group L?group T were lower than group LT(P<0.05) at different time point. The histopathology score of the rat intestine in group L was lower than group T(P<0.05) at different time point. Compare with group N, the levels of MDA?CK in the serum decreased at different time point in group LT?group L?group T(P<0.05), Comparing with group T,the levels of MDA?CK in the serum decreased at different time point in group LT?group(P<0.05). Comparing with group L,the levels of MDA ? CK in the serum decreased at different time point in group LT(P<0.05). Comparing with group N, the levels of ATP in the intestinal tissue decreased at different time point in group LT?group L?group T(P<0.05). Comparing with group T,the levels of ATP in the intestinal tissue decreased at different time point in group LT?group L(P<0.05). Comparing with group L, the levels of ATP in the intestinal tissue decreased at different time point in group LT(P<0.05). And the levels of ATP in the intestinal tissue decreased at different time point in group LT?group L?group N(P<0.05),the levels of MDA?CK in the serum and the levels of ATP in the intestinal tissue in group L was lower than group LT(P<0.05) at different time point.Conclusions 1Trimetazidine may improve the intestinal smooth muscle cells metabolism,so preventing intestinal infarction. 2Low molecular heparin may reduce MVT intestinal damage by improving the microcirculation. 3Low molecular heparin combined trimetazidine synergy for the treatment of MVT, so preventing intestinal infarction wall permeability.
