Effect Of Rhizoma Chuanxiong On Osteoblast Proliferation And TypeⅠCollagen Expression

Objective: Through experiment Hort extract and ligustrazine five groups of different concentration on the effect of bone cell proliferation,alkaline phosphatase and typeⅠ collagen expression,the most ideal effect of the concentration.Provide a reference for clinical application.Method: In the experiment the E1-MC3T3 of mouse osteoblast cell line was used as the research object,the extract of Rhizoma Chuanxiong were divided into five groups with different concentrations of Ligustrazine.The effect of Ligustrazine on the proliferation of osteoblasts after 24 h,48h and 72 h were observed in five groups of different concentrations of Ligustrazine;Enzyme linked immunosorbent assay was used to determine the activity of ALP in 2 days,4 days and 6 days;after 2 days of culture,the expression of typeⅠ collagen protein was detected.Result: The concentration of 1.5.10.15.20ug/ml five groups of concentration of Ligustrazine extract on the proliferation of osteoblasts have promoted the role of.The concentration of 1.5.10.15.20ug/ml five concentration of Ligustrazine can improve the activity of alkaline phosphatase(ALP)in osteoblasts.Comparison of ug/ml 1.5.10 and blank control group(P<0.05),15.20ug/ml and blank control group(P>0.05).The concentration of 1.5.10.15.20ug/ml five groups of concentration of Ligustrazine extract can promote the expression of typeⅠ collagen.Conclusion: This experiment through in vitro cell culture technology,from the level of cell biology the results of the study show that Chinese medicine chuanxiong extract and Ligustrazine on mice into osteoblast MC3T3-E1 proliferation,alkaline phosphatase and typeⅠ collagen has role in promoting,the concentration of 10 ug/ml was the ideal concentration of promoting osteoblast proliferation and typeⅠ collagen expression.This provided the specific reference data for the animal experiments and clinical experiments in the future.
