Objective: Glycyrrhizae Radix et Rhizoma is the dry root and rhizome of Glycyrrhiza uralensis Fisch,G.inflata,G.glabra L.belongs to Leguminosae.At present,there are many researches about the antioxidant effect of Glycyrrhizae Radix Et Rhizoma,however the research of the active ingredient of Glycyrrhizae Radix Et Rhizoma is still at the primary stage.The experiment aims to investigate the role and the mechanism of glycyrrhizic acid and glycyrrhizic flavone in protecting skin photoaging,which lays the theoretical foundation of safe and effective UVB protection drug development.Methods: Photaging model is established by HDF cells after UVB(30m J/cm2)irradiating,different concentration glycyrrhizic acid(10-7mol/L?10-8mol/L?10-9mol/L)and glycyrrhizic flavone(10-9mol/L?10-10mol/L?10-11mol/L)were added to handle photoaging HDF cells.MTT method was applied to test cells activity;Kit method was applied to determine activity of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD),and content of malondialdehyde(MDA).RT-PCR method is applied to test expression content of matrix metalloproteinase-1(MMP-1)and matrix metalloproteinases-3(MMP-3);Western blot method was applied to test content of matrix metalloproteinase-1(MMP-1),matrix metalloproteinases-3(MMP-3)in cells.Results: GSH-Px and SOD activity decreased after HDF cells irradiated by UVB,MDA content increased,MMP-1?MMP-3 m RNA expression content and MMP-1?MMP-3 protein expression level increased(P<0.01,P<0.05).Three kinds of different concentration glycyrrhizic acid and glycyrrhizic flavone were added to compare with model groups,cell activity increased at different levels,MMP-1?MMP-3m RNA in cells and protein content in MMP-1?MMP-3 were decreased by different levels(P<0.01,P<0.05).Conclusion: Glycyrrhizic acid and glycyrrhizic flavone can protect UVB causing HDF cells photoaging,the mechanism relates to oxidative damage inhibition,eliminating oxygenic free radical,MMP-1 and MMP-3 protein synthesis.The thesis investigates the role of glycyrrhizic acid and glycyrrhizic flavone in protecting skin photoaging,which lays the solid foundation of the development of skin photoaging drugs and effective cosmetics from the perspective of theory.
