Objectives:To screen and analyze differently expressed circRNAs in plasma and PBMC of the patients with ESRD due to chronic glomerulonephritis by circRNA expression profile microarray for the further exploration in the pathogenesis,diagnosis and treatment.Methods:The plasma and the PBMC?Peripheral blood mononuclear cell? were obtained from 10 patients with ESRD due to chronic glomerulonephritis and 10 healthy people.The circRNA expression profile in the two groups was detected by the human circRNA microarray.The differently expressed circRNAs were selected. Some specific circRNAs were verified by quantitative real-time PCR.Results:710 differently expressed circRNAs were detected?Fold-change?2.0? in plasma,account for 27.58%?710/2578? of the total circRNAs,in which,385 circRNAs were up-regulated,account for 14.93%,325 circRNAs were down-regulated,account for 12.60%.968 differently expressed circRNAs were detected?Fold-change?2.0? in PBMC,account for 29.24%?968/3310? of the total circRNAs,in which,670 circRNAs circRNAs were up-regulated,account for 20.24%,298 circRNAs were down-regulated,account for 9.00%.12 circRNAs were verified by quantitative real-time PCR,which were closely associated with uremia,such as hsacirc0044922?hsacirc0037409?hsacirc0004951?hsacirc0049271?hsacirc0053958?hsacirc0000369.Validation results were consistent with the results of microarray gene expression.Conclusions:The expression profile of circRNA in patients with ESRD was changed,which implied, changed circRNAsmight be the potential diagnosis biomarkers and played an important role in the occurrence and development of uremic patients.
